Zyxin gene is located at chromosome1of red gallus, and this protein is a kind of phosphoproteins which could comprise Zn and fat, gathering in the adhesion plaque which is rich in actin, moving along the actin skelemin. It have the effects of the movement of the cells, growth and migration of Tumor cells and adjusting mitosis, also working as transcription-activator in cellular in coordination with other proteins. It is a kind of intracellular protein regulating cytoskeleton-actin. Zyxin protein remodels actin cytoskeleton during trauma repairing, migration, adhesion.Epithelial mesenchymal transformation. Studying also shows there is significant association between gene zyxin SNP and NO-index changing after coccidiosis infection, which is the important genetic marker reflecting resistance of coccidiosis in chickens. This study works out the SNP of nine zyxin exons of Jinghai yellow chicken by PCR-SSCP and sequencing (Suntory chicken as the contrast group). Analyzing the population genetic characteristics and the association to related characters by SNP checked out. The results show as follow:Twelve pair primers completely covering nine exons of Zyxin gene were designed and SNPs were detected by PCR, totally4SNPs were found with P1, P3A, P6, P9C. The gene frequencies and genotype frequencies both fit the Hardy-Weinberg equilibrium at P3A, P6loci indicated by the χ2test, which may result of Jinghai Yellow chicken.By the method of PCR-SSCP, four mutation sites were found:At zyxin exon2152C to T which leads to a change in amino acid from Leu to Phe; At exon4of zyxin375T to G which did not lead to a change in amino; At exon7of zyxin130G to T which leads to a change in amino acid from Gly to Cys; and at exon9of zyxin966G to A which leads to a change in amino acid from Gly to Cer.The result of analysis between different genotypes for primer1and growth traits of Jinghai Yellow Chicken showed that:In males, BB genotype was significantly higher than the other two genotypes on the body weight (BW) at4weeks age, abdominal fat, and dressing percentage (P<0.05), as well as superior on BW in the other stages and on slaughter traits. In females, BB genotype was better on BW at8to16weeks.The statistical result of primer3A between different genotypes and chicken growth traits of Jinghai Yellow Chicken showed that:AA genotype was higher than other genotypes on the body weight in both males and females (P<0.05); in males AA genotype was significantly higher than AB and BB on the heart weight (P <0.05) and was superior to most other carcass traits. In females; BB was a kind of advanced genotype, but the difference between genotypes on growth traits was not significant.To primer6, the result analysis of different genotypes and chicken growth traits of Jinghai Yellow Chicken showed that:In both males and females, BB genotype was superior to other genotypes on the body weight from4to16weeks, and from8to12weeks the difference was significant (P<0.05); in males genotype BB was significantly higher than AA (P<0.05) in liver weight and dressing rate, but on the abdomen fat weight, breast meat rate and leg muscle rate, AA was an advanced genotype. BB genotype was superior to AB and BB on body weight in the other stages in males and females. In females most of the carcass traits trends was BB>AA. Above all, the allele B of this site might enhance the body weight and carcass traits in Jinghai Yellow Chicken.The statistical result of P9C between different genotypes and chicken growth traits of Jinghai Yellow Chicken showed that:AA genotype was superior to other genotypes on the body weight in males (P<0.2); In females BB genotype was superior from4to6weeks (P<0.05) but the Later trend was consistent with males. Analysis of carcass traits showed that genotype AA was significantly higher than AB (P<0.05) in breast muscle weight in males. Most other traits AA performance is excellent, but the difference was not significant (P>0.05). In females AA genotype was bigger than AB (P<0.05) on live weight,head weight,foot weight and weight of body carcass(P<0.05).The result of zyxin analysis in Suntory chicken showed that:all sites were not significantly related to the Suntory hens; in males P1genotype BB was significantly higher than AB in head weight and genotype AB was significantly higher than AA in breast muscle rate; P3A genotype AB was significantly higher than AA (P<0.05) in both stomach weight and breast muscle rate; P6genotype AA was significantly higher than AB and BB (P<0.05) in breast muscle rate and genotype AB was significantly higher than AA in leg muscle rate;In males primer9C genotype BB was significantly higher than AB in live weight, liver weight, leg muscle weight and leg weight. The differences between two breeds may be related to insufficient number of experimental groups and it is necessary for further experiments.The result of zyxin meat quality analysis between different genotypes of Jinghai yellow chicken showed that:Compared with other primers, only P1has significant influence on Chest muscle hyden loss rate in Jinghai Yellow chicken males and Chest muscle color in Suntory females; P3was significantly correlated with Jinghai Yellow females and Chest muscle color in Suntory females. Therefore we got the message that zyxin had no affection on meat quality in neither males nor females, which showed zyxin may be a potential molecular marker in disease resistance and slaughter traits.
