Rice blast is one of the most serious diseases of cultivated rice throughout the world. Magnaporthe oryzae(M. oryzae) is a filamentous ascomycete fungus and has a typical life history, and has been developed as a model organism for the investigation of fungus-host interaction.With the accomplishment of Human of Genome Project, the biological research comes to the new post-genome era. Digital gene expression analysis (DEG) is a powerful high-throughput experimental technique designed to gain a qualitative and quantitative measure of global gene expression in eukaryotic organisms. Using the analysis technologies of computer science to analysis the millions data and discover the useful and instructive knowledge of biological experiment is attracting more and more attentions for the information biology.In this paper, we select some genes by the analysis of two temperature-sensitive mutant gene expression, and try to find associated with the pathogenicity. Selected two temperature-sensitive mutants and detected their phenotypic, The mutants showed decreased delayed germination of conidium, the pathogenicity of mutants to barley leaves was also weakened. Through their transcriptional expression profiling sequencing, we get differential expressed genes.Selected11genes randomly, design the primers for qRT-PCR and verify the result from DEG, The results showed that the mutants has the same expression trend of10genes with the Guy-11.The qRT-PCR results and SAGE sequencing results of T-146existence significant correlation, but the T-154does not exist significant correlation. And then construction of vectors for gene replacement and has get some mutants.We get6gene deletion transformants of MGG07200and1randomly inserted transformants of MGG01722.Gene expression profiling analysis is relatively new in recent years, gene expression analysis to study the relation of the genome and pathogenicity is more effective and meaningful.
