The aim of study that is21days chicks infected with IBDV expounds immune suppressionwas caused by IBDV through cell apoptosis. HE, TUNEL, Real time Fluorescent quantitative PCR,IHC and other technical were used for revealing the number of apoptosis cells, the changes ofnuclear cytoplasm ratio, and the dynamic expression of Bcl-2, Bax and p53in immuneorgans-bursa of Fabricius, thymus and spleen. The study aimed at comprehensive and systematicreveal IBDV influenced the number of apoptosis cells and expression of related genes in immuneorgans of SPF chicks. It is supplying a new road to treat IBD.The results show that:1. After21-days-SPF-chicks were infected with IBDV, the number of apoptotic cells in bursaof Fabricius, thymus and spleen of infected chicks were higher than of control chicks in3~7d(P<0.01or P<0.05); the apoptotic cells in bursa of Fabricius is much more than in thymus andspleen. After3~7d which chicks infected virus, the number of nucleus cytoplasm ratio of cells inimmune organs of infected chicks were lower than of control chicks (P<0.01or P<0.05), there areno significant change in other organs(P>0.05).2. After21-days-SPF-chicks were infected with IBDV, the number of Bcl-2positive cells inabove immune organs of infected chicks were outstanding higher than of control chicks afterinfected3~7d (P<0.01); the expression of bcl-2mRNA in three immune organs increased, theexpression of bcl-2mRNA in bursa of Fabricius of infected chicks were much higher than ofcontrol chicks after infected3~7d (P<0.05), and the expression of bcl-2mRNA in thymus ofinfected chicks were very significant higher than of control chicks in3~5d(P<0.01), and theexpression of bcl-2mRNA in spleen of infected chicks were significant higher than of controlchicks in3d(P<0.05). After chicks were infected with IBDV, the number of Bax positive cells inbursa of Fabricius of infected chicks were higher than of control chicks after infected3~7d(P<0.01), and the number of Bax positive cells in thymus and spleen of infected chicks werehigher than of control chicks after infected3~28d(P<0.01and P<0.05); while the expression of baxmRNA in thymus, spleen and bursa of Fabricius of infected chicks were significant or verysignificant higher than of control chicks in3~7d (P<0.01or P<0.05).Bcl-2/Bax ratio in infectedchicks were lower than in control chicks, but there are no significant changes (P>0.05).3. After21-days-SPF-chicks were infected with IBDV, the expression of p53mRNA in thymus and spleen of infected chicks were much higher than of control chicks in3d (P<0.05),lower than of control chicks in the rest of control point, and no significant changes (P>0.05).Theexpression of p53mRNA in bursa of Fabricius of infected chicks were significant and verysignificant higher than of control chicks in5~7d (P<0.01and P<0.05), another time is lower thanof control chicks (P>0.05).
