Drunken horse grass (Achnatherum inebrians) distributed in northwest China’s pasturelands of Gansu, Inner Mongolia, Qinghai, Xinjiang, Tibet and other regions, which is mainly one of the main poisonous grass on native grassland in north China. A. inebrians from Tianzhu county, Gansu province of China was used in this study. Ergot alkaloids contents in different individuals and potted plants under the conditions of different concentrations of Nitrogen (N), Phosphorus (P) and N+P combination, and tryptophan were detected by using high performance liquid chromatography (HPLC) analysis. Hope to find the optimal conditions to produce alkaloids by endophyte-infected A. inebrians. The main results are as follows:1. Detecting an ergot alkaloid-lysergol solution under different period with UV lamp found that lysergol sample peak area decreased along with the increasing of irradiation time. It was found that UV lamp irradiated every30minutes, the peak area was decreased by30%.2. Wild A. inebrians from Dachaigou, Tianzhu, Gansu of China, and potted at greenhouse detection indicated that different individuals of Neotyphodium gansuense infected A. inebrians had different ergot alkaloids contents, which ergine and ergonovine were11.89-908.14mg/kg grass dry weight and0.09-53.63mg/kg grass dry weight, respectively.3. Under treatments with tryptophan, N. gansuense infected A inebrians showed that with the addition of different contents of tryptophan, biomass, plant height and tiller number of the seedings were lower than those of without tryptophan, however, ergine and ergonovine contents were increased significantly (P<0.05).4. Under different concerntrations of N, P and N+P combination, N. gansuense infected A. inebrians found that N, P and N+P treatments on biomass, plant height, tiller number of the grass increased with increasing of N, P concentrations. The maximum concentrations of both ergine and ergonovine appeared under either high N concentration or middle P concentration under only N or P treatments. Furthermore, the maximum concentration of ergine under high N+middle P treatment, and the maximum concentration of ergonovine under high N+high P treatments.
