| Citrus, which is one of the most important fruit tree in the world, contains a plenty of nutrients and bioactive compounds, has a high value of nutrition, pharmaceutical and health protecting function. A systematic study of the scientific use of citrus fruits on their nutrition and health function can not only improve the value of citrus products, but also benefit the high efficiency utilization of the citrus genetic resources and the development of citrus industry. Citrus fruits are not only consumed as fresh product, but are also used in various areas, such as processing, hairdressing, antimicrobial, insecticidal, medicine and health care. Recently, the improvement of metabolic disorders of citrus fruits has received increasing attention. China is one of the earliest and most important origin centers of cultivated citrus in the world. Therefore, researches on the anti-metabolic disorders effects and its mechanisms of the endemic citrus germplasm of China can provide not only theoretical significance for the high efficiency utilization of the genetic resources but also a positive role in promoting the development of citrus industry.The fruits of Citrus ichangensis Swingle and their different parts (peel and pulp) were chosen as the plant materials in this study. The total contents of polyphenol and flavonoids in extracts were detected by spectrophotometry and the contents of flavonoids composition were investigated by HPLC. The effects of citrus fruits extracts on body weight gain, food intake, fasting blood glucose concentration, serum lipid and insulin levels, hepatic and fecal lipids content, glucose tolerance and insulin resistance were investigated in high-fat diet-induced mice. The expression levels of glucose and lipid metabolic-related genes in the liver tissue were analyzed by real-time RT-PCR. The data obtained were examined by Excel and the Figures were created by Photoshop7.0software. The main results are as follows:1. According to the results obtained, the contents of total polyphenol, total flavonoids and the main flavonoids were higher in Citrus ichangensis peel extract compared with the pulp extract.2. Our data showed that the peel extract was more efficiency on anti-obesity compared to the pulp extract, and it lowered fasting blood glucose levels and improved glucose tolerance significantly.3. These results suggested that Citrus ichangensis peel extract could prevent diet-induced obesity independent of food intake inhibition and lipid absorption in the intestine. In the HF+peel group, body weight gain, fasting blood glucose concentration, serum total cholesterol (TC), low density lipoprotein cholesterol (LDL-C) and insulin levels, liver triglyceride (TG) and TC concentrations were significantly (p<0.05) decreased relative to those in the HF group.4. The anti-metabolic disorder effects of Citrus ichangensis peel extract may involve in multi-molecular targets, including the inhibition of PPAR-y and LXRs in the mouse liver. |
