The composition and bioactivity of tea polysaccharides of 140 varieties were studied, relatively higher and lower bioactivity representative breeds were selected. Monosaccharides composition,spectral signature of ultraviolet spectrum and infrared spectrum of the tea polysaccharide were analyzed based on the higher bioactivity representative variety NO.48.1.140 tea varieties were collected,picking standard was a bud with three leaves.The samples were fixed by steam.After water-extraction and being precipitated by ethanol, the tea polysaccharides of different breeds were obtained.The extraction rate of tea polysaccharides and the contents of neutral sugar,uronie acid,protein,polyphenol of each crude polysaccharide were analyzed,and the scavenging rate of·OH,O2-·by tea polysaccharides of 140 varieties were compared.The results showed that:there were very significant differences of extraction rate of tea polysaccharides and the contents of neutral sugar,uronic acid,protein,polyphenol of each crude polysaccharide among 140 breeds,it illustrated that the breeds diversity of tea polysaccharides was clear.Meanwhile,there was a very significant difference of scavenging rate of·OH2 O2-·by tea polysaccharides of different varieties.NO.137,NO.96,NO.108,NO.72,NO.90,NO.41, NO.48,NO.83,NO.56 were selected as the higher bioactivity representative varieties,and NO.21,NO.57,NO.22,NO.117,NO.93,NO.28,NO.50,NO.37,NO.55 were the lower bioactivity representative variety based on cluster analysis of scavenging rate of·OH, O2-·by tea polysaccharides of 140 varieties.2.NO.48,the higher bioactivity representative breed,was selected as research objects.The crude tea polysaccharides was extracted,and four main fractions TPS1,TPS2, TPS3,TPS4 were isolated from crude tea polysaccharides by DEAE-52 cellulose column.TPS2 was separated by Sephadex G-150 column chromatography into TPS2-1. Monosaccharide Composition,molar ratio,spectral signature of ultraviolet spectrum of TPS1,TPS2,TPS3,TPS4,TPS2-1 were analyzed.Gas chromatography analysis showed that TPS1 was composed of mannose,arabinose,glucose,galactose,xylose, monosaccharide molar ratio was:3.1:1.55:3.57:9.14:0.80;TPS2 was composed of rhamnose,mannose,arabinose,glucose,galactose,xylose,and monosaccharide molar ratio was:0.45:3.19:0.39:0.19:1.28:4.56;TPS3 and TPS4 were both composed of rhamnose,rhamnose,arabinose,glucose,galactose,monosaccharide molar ratio were: 0.36:1.11:0.31:2.28 and 0.40:0.61:0.42:1.93;TPS2-1 was composed of rhamnose,arabinose,xylose,mannose,glucose,galactose,monosaccharide molar ratio was:0.64:4.43:0.48:0.28:1.47:5.82.UV and IR spectrum analysis showed that the tea polysaccharides was a polysaccharide-protein conjugate withβ-pyran.
