Since1940s, fungi polysaccharides were found to possess anti-cancer activity,domestic and foreign researches on natural bioactivity of polysaccharides have beenpaid more and more attention. This paper optimized the extraction and purificationprocess of chestnut polysaccharide, compared the infrared spectra of polysaccharideand composition and content of monosaccharide among different chestnut varieties(lines), investigated the structure and antitumor activity of chestnut polysaccharidesusing Yanshan chestnut as test materials. These have great significance toscientifically evaluate the utilization value of chestnut polysaccharide, preferablyexploit and use chestnut germplasm resources. The main research contents and resultsare as follows:1.The optimum extraction technology of Chinese chestnut polysaccharide. Thetechnology is tested with optimal methods like hot water-assisted extraction,microwave-assisted extraction, hydrochloric acid-assisted extraction andultrasound-assisted extraction polysaccharides. Compare the extraction time andsolvent usage, calculate the extraction rate, and then come to a conclusion of what isthe best chestnut polysaccharides extraction technology. The results show that theautomated pressurized liquid extraction is a method for extracting polysaccharide fast,less solvent consumption, extraction efficiency.2.Purification and molecular weight studies on the polysaccharide content. Theremoval of protein by Sevage method, then using macroporous resin AB-8to proteinand color, it can make the protein removal rate reached80%, the decolorization ratecan reach86.36%, significantly better than activated carbon decolorization rate andhydrogen peroxide. Purified by DEAE-52cellulose column chromatography andSephadex G-100gel column chromatography for further purification after UVspectrophotometric detection, only one peak, showed that the chestnut polysaccharidewas pure and homogeneous component; the determination of chestnut polysaccharide,weight average molecular weight of3.28×105Da.3.Using IR and GC-MS, analysis of polysaccharide structure, monosaccharidecomposition.The reference to the used of APLE extraction from20different varieties(lines) chestnut polysaccharide, IR showed all chestnut polysaccharide has typicalabsorption peaks of polysaccharides. The Yanshanduanzhi, Dabanhong, Zunyu, Yanlong, Yanwang, Daomazhai, Wangchanggou, Qianxizaohong, Qianxiwanhong,Anyou, Qianxi1, Qianxi5and Qinglong30, has characteristic absorption peak, Wegenerally think of polysaccharide containing characteristic absorption peak hasstrong activity.4.Study on the monosaccharide composition. The GC-MS analysis resultsshowed that the monosaccharide6varieties of chestnut Yanshanduanzhi, Dabanhong,Yanlong, Yanwang, Qianxizaohong and Qianxiwanhong, are composed of ribose,rhamnose, arabinose, xylose, mannose, glucose and galactose. But ‘Yanshanduanzhi’glucose content was the highest, the content of polysaccharide was76.64%,‘Yanwang’ galactose content was the highest, the content of polysaccharide was44.61%.5.The antitumor activity of chestnut polysaccharide was evaluated using‘Yanlong’ polysaccharide as test materials. Chestnut polysaccharide can inhibit humanhepatoma cell Bel-7402, lung cancer cell A-549, uterine cancer cell HCT-8. Theinhibitory rate increased with the concentration of polysaccharide, when theconcentration of polysaccharide was300μg/mL, on human hepatocellular carcinomacell Bel-7402, lung cancer cell A-549, uterine cancer HCT-8cell inhibition rate isrespectively69.54%,51.38%and55.27%, indicating that contains-glycosidic bond‘Yanlong’ had good antitumour activity.
