In order to improve thermal stability and mechanical properties of the grass carp scale collagen-collagen-chitosan blend films and sponges, the natural polyphenols PA (procyanidins), TP (tea polyphenols) was used as crosslinking agent. The mechanical properties, water vapor permeability and other characteristic of the crosslinked films and crosslinked sponge were measured. The structure changes of the films and sponge was characterized by infrared spectrum, X-ray diffraction, differential scanning calorimetry and microscope. Preliminary study on the biocompatibility and cell toxicity of the films and sponge were also includedThe main results were as follows:1. Preparation and characterization of grass carp scale collagen/chitosan blend filmGrass carp Scale Collagen/Chitosan blend films were prepared by solvent-casting method with grass carp scale collagen and chitosan. The results showed that the optimal ratio of collagen and chitosan for preparation of the blend film was 1.5% of chitosan and 0.3% collagen solution mixed at the ratio of 1:3(v/v). The mechanical property and water vapor permeability coefficient of the blend film have been improved because of the strong interaction and good compatibility between collagen protein and chitosan.2. Preparation and characterization of grass carp scale collagen/chitosan blend film crosslinked by natural polyphenols (PA orTP)We found that the crosslinked blend films have beteer swelling and tensile strength when proanthocyanidin (PA) concentration was 0.3%, and TP concentration was 0.5%. Water stability and thermal stability of the crosslinked blend films made in the condition above are improved greatly. DSC analysis showed that, the denaturation temperatures of PA and TP crosslinked blend film was improved to 93.96℃and 97.84℃, separately.3. Morphology and structure analysis of the blend films crosslinked by natural polyphenols (PA or TP)The structural changes of the crosslinked blend films were characterized by optical transmittance, infrared spectrum, X-ray diffraction, differential scanning calorimetry microscope, SEM, and FT-IR methods. The result showed that polyphenols, collagen and chitosan has strong interaction and good compatibility.4. Biological compatibility of the blend films crosslinked by natural polyphenols (PA orTP)The crosslinked blend films has very good biocompatibility, the hemolytic hemolysis of the crosslinked blend films is far less than the standard (5%).5. Preparation and characterization of grass carp scale collagen/chitosan sponge crosslinked by natural polyphenols (PA or TP)The results showed that the best crosslinking conditions of PA was as follows:0.5% C and 0.5% CTS were mixed at the ratio of 2:8(v:v), adding 0.5% PA as crosslinking reagent with pH 7.4 40% ethanol PBS solution as media using bath method. And the best crosslinking conditions of TP was as follows:0.5% C and 0.5% CTS were mixed at the ratio of 2:8(v:v), adding 0.5% TP as as crosslinking reagent, crosslinked in pH 7.4 40% ethanol PBS solution using bath method. The water stability and thermal stability of the crosslinked sponge made in the optimal condition were improved greatly. DSC analysis shows that, the denaturation temperatures of PA-crosslinked sponge was improved to 87.61℃, the denaturation temperatures of TP crosslinked blend film was improved to 94.91℃.6. Biological compatibility of the crosslinked sponge crosslinked by natural polyphenols (PA or TP)The crosslinked sponge has very good biocompatibility, the hemolytic hemolysis of the crosslinked blend films is far less than the standard (5%). Compared with gelatin sponge, The crosslinked sponge has good hemostasis performance.
