Collagen is the commonly used clinical material for haemostasis, which has the haemostasis effect by inducing platelet aggregation, promoting thrombogenesis and activating the blood clotting system. Chitosan is a widely used medical high polymer material in medical treatment at present because of its security and reliability. At the same time, it has the superordinary bioaffinity and mechanics characteristics. Our research has produced human-like collagen by recombinating E coli BL21 high density fermental technology. Then, we use human-like collagen as the main material, reunioning the specific biont polysaccharides, to make chitosan-human-like collagen bibulous by cryodesiccation technology. Through cell and animal experiment, we expect to confirm that chitosan-human-like collagen bibulous has better hematischesis effect than other common hematischesis materials in clinical at present (gelatin sponge,e.g.), because it has the bionomics of both human-like collagen and chitosan. Moreover, chitosan-human-like collagen bibulous has preferably biocompatibility.The physics and chemistry nature of chitosan-human-like collagen bibulous were observed. chitosan-human-like collagen sponge was a kind of new hemostatic material, its outward appearance is the faint yellow sponge, the internal aperture probably is 100 ~ 250μm, its porosity achieves above 90% and water-absorptivity probably is 3100%, the density is 0.043 g/ cm3, the ash is less than 0.1%. The sponge was put in the physiological saline in 8 weeks, it does not damage and the shape integrity, so chitosan-human-like collagen sponge has the good physics and chemistry nature. Our research about the biocompatibility of chitosan-human-like collagen bibulous includes: blood compatibility test, cytotoxicity test, sensibilization test, partis stimulation test and carcinogenicity test. Experimental result shows:①chitosan-human-like collagen bibulous does not induce hemolytic reaction, it has no effect on the function of red blood cells, white blood cells and blood plate, it has satisfactory blood compatibility. (2) chitosan-human-like collagen bibulous does not damage the morphous and the growth of human collagenoblast by cell morphous method and MTT chromatometry.③chitosan-human-like collagen bibulous does not produce sensibilization reaction to the skin of cavia cobaya, and does not have any provocation to the skin of rabbit;④chitosan-human-like collagen and gelatin sponge were buryed in the liver raw surface of rabbits, which were sacrificed after 15 days. Gelatin sponge cause more severe tissue and cell changes compared to chitosan-human-like collagen through observation of light microscope and electron microscope.⑤The degrading rates of chitosan-human-like collagen and conventional material did not have significant variance.?subcutaneously embedding chitosan-human-like collagen bibulous does not have any carcinogenicity and does not have toxic effect on the blood, hepatic function and renal function. Therefore, chitosan-human-like collagen bibulous has favourable biocompatibility and blood compatibility.We have deeply researched the hematischesis effect of chitosan-human-like collagen bibulous on liver raw surface of rabbit. we make the raw surface which is about 1cm×1cm on the liver of rabbit, then we use chitosan-human-like collagen bibulous and gelatin sponge to stop bleeding. Negative control group is to use the medical carbasus parties hemostasis with packs after making the raw surface on the liver of rabbit. We find the material which has the best hematischesis effect by bleeding time and bleeding volume measure. The result shows that chitosan-human-like collagen bibulous and gelatin sponge have significant difference on the bleeding time and bleeding volume (P<0.05). Accordingly, we consider that chitosan-human-like collagen bibulous has better hematischesis effect compare to gelatin sponge.
