Spectrum of serum peptides are analysised by mass spectrometry which purpose is toobtain accurate peptides’s molecular.In order to find disease marker, further analysis ifpeptides sequences by biological mass spectrometry.Early detection can significantly improvepatient survival, reduce patient mortality, so early diagnosis is very important for the patient’sprognosis.This paper studies efficient and stable in serum samples of enriched peptidecomposition, and use of the WCX (waek cation exchage) magnetic system and massspectrometry detection of ischemic cardiomyopathy patients with heart failure serum peptidemap.Through bioinformatics software to analyze the peak of its characteristic signs,and findthe peptide map for diagnosising ischemic cardiomyopathy patients with heart failure.all theseresearch is to establish the methord for diagnosis and antidiastole in ischemic cardiomyopathywith heart failure patients.【Objective】The present study provided the basis for the diagnosis of IschemicCardiomyopathy patients, and determined the specific serum peptide profile by comparingthe serum differences between Ischemic Cardiomyopathy (ICM) patientsand normal controlgroup. In addition, this study also discuss the function of the difference protein.【Methods】 The study collected pre-treatment of ICM and normal control serumsamples,which were followed by automated MALDI-TOF MS analysis after peptidesextracted on magnetic beads coated with WCX phase. Mass spectrographic data was analyzedwith ClinproToolTMsoftware,such as GA、SNN and QC. The specific serum peptide modelof ICM was established by using genetic algorithms.【Results】 Through extracting by bead technology with mass spectrometry healthy anddisease serum, this study can detect139serum peptides. The study detected IschemicCardiomyopathy serum peptidome patterns. The differential expression of the peptide peaksare47(n=47), the study selected1866.17Da、2660.54Da、7761.49Da、1848.81Da、 2037.57Da peptide peaks,which were statistically significant differences between the twogroups, to establish Ischemic Cardiomyopathy diagnostic model; The1866.17Da、7761.49Da、1848.81Da、2037.57Da low expression in the disease group;2600.54Da highexpression in the disease group.The recognition rate, predictive power, sensitivity andspecificity of the model were100%and97.49%. One significant difference between the peaksidentified by tandem mass spectrometry is the HSP60protein fragments,it can impedecardiac apoptosis,protect ischemic myocardial cell microtubules and actin cytoskeleton。【Conclusions】The specific serum peptide model had certain applications in thediagnosis and antidiastole of nasopharyngeal carcinoma with heart failure, and provided thebasis for discovering a specific marker of Ischemic Cardiomyopathy.
