The Experimental Study On Parecoxib Sodium Combined With Methyl-Prednisolone For The Repair Of Sciatic Nerve Injury In Rat

Objective: To compare a single injection parecoxib sodium, methylprednisoloneand parecoxib sodium combined with methylprednisolone for the repair of rat sciaticnerve injury, and to provide treatment reference for the patients who underwentperipheral nerve damage clinically.Methods: We selected forty Sprague-Dawley (S-D) rats, weighed180-200g,male or female rats, randomly divided into four groups (n=10each). Group C, thecontrol group; P group: the parecoxib sodium group; M group: methylprednisolonegroup; P+M group: parecoxib sodium+methylprednisolone group. With pentobarb-ital sodium anesthesia and under sterile operation, we clamped right sciatic nerve ofrats so that it causeed crush injury with Sunderland Ⅱ degree. The groups of experim-ent rats were given parecoxib sodium20mg/kg, methylprednisolone20mg/kg, parec-oxib sodium20mg/kg combined with methylprednisolone20mg/kg, respectively byintraperitoneal injection after operation one hour. The control group were only sepa-rated the right sciatic nerve and clamped them, but they were not given any drugs, andthe others as well as the experimental groups. We estimated the sciatic function indexafter the first week, the second week, the third week, and the sixth week and the gast-rocnemius wet weight rate after the sixth week, respectively.Results:1. There was no infection or autotomy in every group rats after operation.2. Except the sixth week, compared with group C, the SFI was statistically signific-ant in the P group, M group and P+M group after surgery in the first week, thesecond week and the third week（P＜0.05）; it was not statistically significantamong P group, M group, and P+M group.3. The gastrocnemius wet weight rate was statistically significant between P groupand C group（P＜0.05）,and it was not statistically significant among other groups（P＜0.05） in the sixth week. Conclusion:Both parecoxib sodium and methylprednisolone could promote the repair of sciaticnerve injury in rats；it was not superior to inject two drugs than one drug separately.
