Objective:To observate effection of simvastatin on asthma rats modelserum Interferon-γ、 Interleukin-4level change,so as to provide theexperimental basis of simvastatin in treatment of bronchial asthma.Methods: Take healthy SD rats,male,a total of thirty-two,were randomlydivided into four groups,respectively: normal control group,asthmaticmodel group,dexamethasone group and simvastatin group,with eight ratsin each group.By ovalbumin (OVA) induced by intraperitoneal injection ofsensitivity and atomizing inhalation of excitation preparationeal of ratasthma model,with simvastatin orally,and compared with normal controlgroup、model group and the dexamethasone group.With two-site sandwichensyme linked immunosorbent assay to detect the rat serum IL-4andIFN-γ content.HE staining were observed under microscope rat rightpulmonary pathological changes of lung tissues.Results: Model group serum IFN-γ content was significantly lower thanthat in the control group,IL-4content was significantly higher than thosein the normal control group,there was significant difference (P <0.01),respectively (83.71+/-8.89) pg/ml and (106.62+/-13.33) pg/ml;Comparedwith the model group,simvastatin group and the dexamethasone groupserum concentration of IFN-γ were (95.93+/-11.74) pg/ml and(96.34+/-11.64) pg/ml,significant higher than the asthma model group,there was significant difference (P <0.05), and between the two treatmentgroups didn’t significant difference (P>0.05);To compared with themodel group, two of IL-4in treatment group was obviously decreased (P <0.01), the content of IL-4were (79.08+/-7.34)pg/ml and (73.39+/-7.76)pg/ml, drug treatment group had no difference (P>0.05).Pathological changes in the model group is visible in the bronchialwall、bronchial and vascular surrounded by a large number of eosinophils、lymphocytes and inflammatory cell infiltration,bronchial mucosal edema,thickening of the smooth muscle layers,airway epithelial cell necrosis, microvascular leakage,within the lumen of a large number of discharge.Were given simvastatin and after treatment with dexamethasone,visibleinflammatory cell infiltration reduce bronchial mucosal edema,reduce ordissipated,airway epithelial cells necrosis loss reduction, intraluminalbronchial secretions decreased or disappeared.Conclusion: On airway inflammation in asthma,simvastatin anddexamethasone, plays anti-inflammatory effects,significantly increase inasthma rat serum levels of IFN-γ,lower serum IL-4level,for asthma whichhave relatively good effect in the prevention and treatment.Themechanism of action may be regulated through the levels of IFN-γ、IL-4,corrected Th1/Th2cell imbalance, inhibited airway inflammation inasthma and relieve asthma.
