Objective To investigate the expression of microRNA-183(miR-183)ingastric cancer (stage Ⅱ GC) and its association with theclinicopathologic features of GC and Ezrin expression.Methods Specimens of stageⅡGC and normal tissues more than5cm awayfrom the tumor tissues were collected from72patients. SYBR Greenreal-time RT-PCR was used to detecte the miR-183expression,Immunohistochemistry was used to examine the Ezrin protein expression inthe tumor tissue. The association of miR-183expression with theclinicopathologic features of stage Ⅱ GC and Ezrin expression wereanalyzed.Results By real time RT-PCR test,miR-183could be detect in paraffin-embedded stage gastric cancer tissue. Compared with the non-tumor controlsamples,the median of relative expression of miR-183was0.676(25th-75thpercentile,0.330-1.253) in the tumor samples,significantly lower thanthat of the non-tumor control samples(set at1.000, P<0.05).The lowerexpression of miR-183was positively correlated with histologicaldifferentiation(0.429vs0.907,P<0.05),lymph node positivity(0.507vs0.908,P<0.05)and shorter survival of patients(56.8%vs80.0%,P<0.05). The expression level of Ezrin in gastric cancer were significantlyhigher than that in the normal tissues,there was a negative correlationbetween the expression of miR-183and Ezrin in stageⅡGC tissues(r=-0.272,P<0.05).Conclusion These results indicate that the miR-183probably involved instageⅡGC infiltrate、metastasis and may become one of the indicator of prognosis,Ezrin is possibly one of targets of miR-183....
