Objective: Different methods were used to detect the expression of chemokines andreceptors in the tumor and liver tissue from patients with the Primary HepatocellularCarcinoma and analysis the relationship with the clinical and pathological features ofthe Primary Hepatocellular Carcinoma. Method:1. Collect the tumor tissue and tissueadjacent to tumo(rThe liver tissue is away from the tumor edge1-3cm)from patientswith the Primary Hepatocellular Carcinoma. Fresh homogenate were prepared fromtissues and the supernatant was collected. Suspend the sediment and the lymphocyteswere isolated by lymphocyte separate liquid. Part of the tissues was processed byformalin for paraffin cuts.2. Detect the positive expression of chemokine andchemokine receptor in the paraffin cuts by immunohistochemical method.3. Detectthe positive expression of chemokine and chemokine receptor in the lymphocyte byflow cytometry.4. Detect the positive expression of chemokine and chemokinereceptor in the supernatant by ELISA essay.5. Analysis of all the data about thepatients, comparing the expression differences about chemokine and receptor betweenpatients with different clinical features.Results:1. The expression of IP-10, MCP-1, IL-8, RANTES, CXCR4, CXCR1,CCR5in the tumor tissue, detecting by immunohistochemical method, were44.1%,64.7%,41.2%,47.1%,76.5%,82.4%and70.6%while the expression in tissueadjacent to tumor were20.6%,41.2%,17.6%,23.5%,52.9%,58.8%,47.1%. Thepositive rate in tumor tissue was visible higher than the tissue adjacent to tumor.Among them, CXCR1、CCR5presented in the cytoplasm, CXCR4present both in thecytoplasm and cell membrane, but mainly in the cytoplasm. The expression of IP-10,MCP-1, IL-8, RANTES were also in the cytoplasm. The chemokine receptorsexpression was strength than that of chemokine.2.The results ofImmunohistochemical show that the positive expression of chemokine and factor in tumor tissue was significantly related to whether the tumor have an intact menmbraneand presence of vascular invasion.3. The CXCR4, IP-10and MCP-1positiveexpressed in the lymphocytes isolated from the tumor tissues were higher than thatfrom tissues adjacent to tumor (P <0.05). But the IL-8, RANTES, CXCR1and CCR5expression of tumor tissue were below the tissues adjacent to tumor group.(P <0.05).4. The result of detecting the supernatant by ELISA assay was the same to that ofFCM. ELISA results which show that IP-10, MCP-1expression in tumor greater thanthat in tissues adjacent to tumor, but only the IP-10has statistically significant. On thecontrast, the expression of IL-8in tumor group was less than tissues adjacent to tumorgroup.Conclusions:Three different methods were employed to detect the expression ofchemokine and receptor in tumor tisssue and tissue adjacet to tumor as well as theability of chemokine secreting by lymphocytes. The result suggested that there weresignifcantly differences between tumor group and the tissue adjacent to tumor group.And the expression was related to whether the tumor has an intact menmbrane andpresence of vascular invasion. All this show that chemokine and receptor mayparticipated in the development of PHC by influencing its pathologic process, theinvasion and metastasis etc.
