More and more evidence had evidenced that reactive oxygen species havevery close relationship with hypertension, diabetes mellitus, cataracts and otherdiseases. A large number of studies have also found that antioxidantion mayreduce or potential decrease the symptoms of these disease.Our study was divided into two parts to research the antioxidative of thePanax notoginseng’s extract and Electrolyzed-reduced water on experimentalmodels. The antioxidant role of thirty-seven ethanol extract andElectrolyzed-reduced water system, to explore the mechanism of action anddose-response relationship.The models as follows.Establish oxidative damage model in vivo or ex vivo to study theantioxidation of the Panax notoginseng’s extract and Electrolyzed-reducedwater.Study the dose-effect relationship or machanism by microsomal lipidoxidative damage models induced by three different sources of stinulators: Fe2+/ascorbate,cumene hydroperoxide(CHP),and CCI4/reduced form ofnicotinamide-adenine dinucleotide phosphate(NADPH) and thechemiluminescence model.Cellular oxidative stress models in QZG cells (human liver cell line) andHepG2cells (human liver tumor cell lines).The Electrolyzed-reduced waterallocation medium were used to cultured cells as differrent groups.The oxidativestress injury was caused by tert-butyl hydrogen peroxide (tBHP) through themedium.The oxidative stress injury models in vivo induced by irradiated of60Co-γray, D-galactose and over-drinking ethanol. The Panax notoginseng’s extract orElectrolyzed-reduced water were used to these models and researched theirprotection to those distroy.(1) Irradiated ainimals model.The rats are divided into5groups, such as thenegative control group (NC), positive control group (PC), The Panaxnotoginseng’s extract (50mg/kg d, L),(225mg/kg d, M) and (450mg/kg d,H)(n=8). Experimental animals were expoursed in environment as60Co-γ Raycumulative6Gy dose except the negative control group.The Panaxnotoginseng’s extract were drug in order to treatment the injury, meanwhile therats of positive control group and the negative control group were fed saline asthe same dose of other rats.we found irradiation could induce freeradical-mediated oxidative stress injury reaction noticead,which was marked bythe increase of MDA and GSSG,decrease of GSH and reduction of SOD.Givingdifferent dose of AE-PNG could prevent SD rats as decrease of MDA andGSSG,increase of GSH and CAT、SOD activities.Espesicaly,the high dose ofPNG showed the dicitnct effect on improving various incicators.(2) D-galactose oxidative stress model.32rats are divided into4groups, aswater (W), The Electrolyzed-reduced water Group (ERW), the D-galactose tap model group (the WD) and D-galactose with The Electrolyzed-reduced watergroup (ERW-D). The animals of ERW Aand ERW-D groups were feed TheElectrolyzed-reduced water,and the models were injected the D-galactose100mg/kg.d for2months. The control group was injected with the saline.Allrats were saccrificed at a predetermined time of4months.The livers and bloodserum were prepared for testing MDA、CAT、SODandGST activity.The resultsshowed that the D-galactose could increase oxidative level in the rats wholebody.Blood serum seemed to be more sensitive than liver After feeding theERW,decrease of MDA and increase of antioxidant enmymes activity wereobserved in serum.However,no obvious changes of liver.Meanwhile,D-galactose could increase CAT, Gpx, of GST activity and T-AOC in serum.After feeding the ERW all the antioxidative indexts rduced.(3) The ethanol oxidation stress model:32rats are divided into4groups, fedwith the tap water (W), ethanol with tap water (W-乙), ethanol with TheElectrolyzed-reduced water (ERW-乙) and The Electrolyzed-reduced water(ERW)(n=8). Water group were feed tap water, and the others were feed TheElectrolyzed-reduced water with4months.Then,the model groups were given50%ethanol (0.76ml/100g d) by gavage3days, the control group received thesame dose of saline. All rats were saccrificed at a predetermined time of4months.The livers and blood serum were prepared for testing MDA、CAT、SODandGST activity. The results showed that the alcohol could increaseoxidative level in the rats whole body as MDA,decrease theSOD and Gpx activity in liver. After giving the ERW increase theMDA in the serum drinking water electrolysis inrats given alcohol stimulate the liver MDA content increased, serum T-theAOC, Gpx is also higher than the negative controlgroup, the liver tissue decreased. All the results demonstrated the Panax notoginseng’s extract wasdemonstrated to possess multi-level anti-oxidative stress property and havedose-effect relation both in vitro and in vivo experiments models. Theelectrolyzed-reduced water can effectively improve the body’s anti-oxidativestress activiation,increased some antioxidant enzymes levels,if it was drink alllong it may be change the condition of some disease as senescence,cardiovascular and cerebrovascular and so on.or prevent and adjuvant therapysome clinicl diseases.
