Objective To study the expression of PTEN,Livin in human clear cell renalcarcinomas(Rccc)and to investigate their relationship.Methods From2010to2011,we were collect50examples paraffin waxspecimen of clear cell renal carcinomas as a group were collected in first affiliatedhospital of henan scientific and technical university.It included30males and20females.Aged from36to74years. The organisation cell carcinoma,as konw Fuhrmanwas divided into third-level: Ⅰ level (n=9), Ⅱ level (n=24), Ⅲ level (n=17). Thepathology of clinical,the Robson method were divided into four groups,includingⅠlevel (n=10),Ⅱlevel (n=28) and Ⅲ~Ⅳlevel (n=12).And select the same period25cases of para carcinoma and10cases of normal renal tissues as contrast group.In theseexamples included17lymphatic metastasis.All patients were not in the preoperativeradiotherapy or chemotherapy.Immunohistochemical SP method detected50cases ofrenal cell carcinoma,25cases of adjacent renal tissue, and10cases of normal renaltissue, expression of PTEN and Livin in different grading and staging of renal clearcell carcinoma compared and correlation analysis.Application SPPS17.0statisticalsoftware for all data and chi-square test results,P<0.05indicated significant difference,P<0.01indicated very significant difference.Results The positive rate of PTEN in renal cell carcinoma was significantlylower than the cancer, paraneoplastic and normal kidney tissue, the difference wasstatistically significant(P<0.01).The positive expression rate of PTEN in renal clearcell carcinoma is40.0%(20cases).In adjacent tissue, the positive expression rate of88.0%(22cases) and normal kidney tissue of all the positive expression of100.0%(10cases).The positive expression rates in cancer tissues and cancer, paraneoplastic andnormal tissue were significantly difference(χ~2=23.36, P<0.01).Livin no expression innormal kidney tissue and adjacent tissues, high expression in renal cell carcinoma,there was significant difference(P<0.01)Livin in renal cell carcinoma and the positive expression rate was44.0%, while no expression in adjacent tissues and normal kidneytissue.The positive expression rates in cancer tissues and cancer, paraneoplastic andnormal tissue were significantly difference(χ~2=20.78,P<0.01).In different clinicalstages of renal clear cell carcinoma tissue PTEN expression and Livin there aresignificant differences in line (P <0.01); and the expression of the two factors was asignificant negative correlation (r=-0.547,P <0.01).Conclusion1.The positive expression rate of PTEN in renal cell carcinoma was significantlylower than the cancer, paraneoplastic and normal kidney tissue,compared thedifference was statistically significant(P <0.01).2.The Livin positive expression rate in renal clear cell carcinoma was signi-ficantly higher than cancer, paraneoplastic and normal kidney tissue, compared to thedifference statistically significant(P <0.01).3.Renal clear cell carcinoma, expression of PTEN protein level decreased, andLivin expression level increased significantly.The PTEN and Livin a negativecorrelation, play an important role in the development of renal cell carcinoma, twofactors combined may be used as an important indicator to determine the degree ofmalignancy of clear cell renal cell carcinoma. And can help determine the biologicalbehavior of renal cell carcinoma.
