Objective To study the significance of Vaspin mRNA and Adiponectin mRNAexpression in obese rats and diabetic rat models and analysis the role of them in obesity andglucolipid metabolism. Providing a new thinking and scientific basis for the disease.Methods One hundred male Wistar rats were accommodated to feed with normal chowfor a week and then divided into three groups randomly:the normal control group(NCgroup,n=15) fed with normal chow,the diet-induced obese group(DIO group,n=35) fed withhigh-fat diet,the diabetic group(DM group,n=50) were fed with high-fat diet for4weeks andthen injected strptozotocin(STZ) into abdominal cavityto inducing to diabetes model,thenormal control group were injected equivalent sodium chloride physiological solution. Allrats were single cage breeding, feeding freedom taken water, The animal room temperaturewas (22±5)℃, relative humidity was (50±10)%, light and dark cycle when12h:12h. Allrats were fed with relative fodders for28weeks.In the end of28weeks,overnight fast for12hours in time, Narcotizing the rats by injecting urethane (0.4g/kg) into abdominalcavityto.Opening the enterocoelia in sterile state and removing liver tissues and repacked in1.5ml freeze-stored tube, frozen in liquid nitrogen and then-80℃refrigerator storage.Expressions of vaspin mRNA and adiponectin mRNA in liver tissue of the3groups wereinvestigated by reverse transcriptase polymerase chain reaction(RT-PCR).Results1.The weight and blood sugar were accordance of the three group before experiment (P>0.05). The weight was higher in obesity group and diabetes group compared with controlgroup(P<0.05), while the wheight of diabetes group was lower than obesity group(P<0.05).The blood sugar was higher in diabetes group compared with control group andobesity group(P<0.05). The blood sugar in obesity group is no obvious increase comparedwith control group(P>0.05).2.The expression level of vaspin mRNA in the liver tissue was higher in obesity group anddiabetes group compared with control group(P<0.05), vaspin mRNA was positivelycorrelated with weight(P<0.05). The expression level of adiponectin mRNA was lower inobesity group and diabetes group compared with control group(P<0.05),and negatively correlated with weight and blood sugar(P<0.05).The expression level of vaspin mRNA andadiponectin mRNA in the liver tissue was no statistically significant between the obese anddiabetic grou(pP>0.05). The expression level of vaspin mRNA and adiponectin mRNA in theliver tissue was negatively correlated (P<0.05).Conclusion The expression of Vaspin and Adiponectin mRNA in obese and diabeticrats that may be an adaptation changes of the glucolipid metabolism.The expression of Vaspin and Adiponectin mRNA in obese and diabetic rats were similar tothe expression of them in adipose tissue. The change of expression reflects the seriousness ofglucolipid metabolism disorder.It may be a kompensatorisch results to glucolipid metabolismdisorder.
