Objective:This study was to investigate the effect of small interfering RNA (siRNA)mediated gene silencing of HER2/neu on proliferation and Apoptosis inBladderCancer cell Line BIU-87. And provide the theory foundation of newtreatments for bladder cancerMethods:(1) HER2/neu siRNA was transfected into BIU-87cells;Were divided intoHER2/neu siRNA group,empty liposome group,negative sequence group anduntransfected control group.(2) The proliferation of BIU-87cells was assessed by CCK-8assay.(3) Apoptosis were analyzed by FCM.(4)The mRNA levels of HER2/neu were detected by RT-PCR.(5) The protein levels of HER2/neu were detected by Western-blot.Results:(1) BIU-87cells transfected with HER2/neu siRNA showed a significant decrease ofcell viability from (82.37±0.90)%to (56.76±1.70)%, was statisticallysignificant (P<0.05).At the same time can induce the apoptosis. The apoptosis rateof transfected ground was (45.60±0.70)%,which was statistically differentfrom that of the control grou(3.87±0.30)%,empty liposome group(5.50±0.36)%, negative sequence group(5.33±0.51)%(P<0.05);(2) RT-PCR method to detect HER2/neu gene expression of each BIU-87cell lines.Show that HER2/neu mRNA expression level of BIU-87cells was decreased aftertransfecting HER2/neu siRNA72h (0.26±0.02), and which was statisticallydifferent from that of the control grou(0.73±0.03),empty liposome group(0.71 ±0.02), negative sequence group(0.70±0.01)(P<0.05). There was not statisticallysignificant between untransfected control group, empty liposome group, negativesequence (P>0.05);Western blot results show that negative sequence group, theempty liposome group,and the control group protein expression respectively was(73.85±0.01)%、(76.86±0.01)%、(77.41±0.02)%.HER2/neu proteinexpression of empty liposome group and negative sequence group are notsignificantly inhibited, and with untransfected control group was no significantdifference(P>0.05). HER2/neu protein expression level of The transfected groupwas(49.76±0.01)%,and HER2/neu protein expression level was obviouslyreduce compare to the rest of the three groups(P<0.05).Conclusion:(1) The BIU-87cell survival rate was dropped significantly, and appearing cellsapoptosis after transfecting HER2/neu siRNA. Show that siRNA can producesignificant apoptosis to HER2/neu positive expression bladder cancer BIU-87celllines.(2) HER2/neu siRNA can effectively reduce the HER2/neu mRNA expressions of thebladder cancer BIU-87cell lines, and make corresponding protein expression reduced.There was good specificity for siRNA...
