objective To apply the small interfering RNAs(siRNA) targeting Survivin to inhibit expression of endogenous Survivin gene in bladder cancer cell BIU-87 and to increase the apoptosis of BIU-87 cell.Methods The recombinant plasmid pshRNA-survivin-1, pshRNA-survivin-2 and pshRNA-survivin-3 were constructed and transfected into BIU-87 cell, respectively. The expression level of Survivin was determined by immunohistochemistry. The proliferation of BIU-87 cell was detected by MTT method. The apoptosis in BIU-87 cell was detected by flow cytometry analysis(FCM).Results After injection of recombinant Survivin shRNA plasmid. RNA interference was able to suppress Survivin expression greatly and then inhibited cell growth effectively and induced apoptosis of BIU-87 cell efficiently. The proliferation of BIU-87 cell which was detected by MTT method both decrease distinctively. The apoptosis in BIU-87 cell line were detected by FCM with apoptosis rates at 49.48±1.88%(P<0. 01), 51.37±3.55%(P<0.01) and 50.07±3.95%(P<0.01), respectively.Conclusions Survivin plays a major role in occurrence and progress of Bladder cancer. Specific inhibition of Survivin by RNAi Can induce apoptosis in BIU-87 cell and suppress tumor growth, and should be a new strategy in gene silence therapy targeting survivin.
