The HER2/neu Expression In Renal Cell Carcinoma And Its Significance

Objective:1,To study the HER2/neu protein expression in renal cell carcinoma (RCC) and normal tissues；2,To study the relationship between the HER2/neu protein expression and the TNM stage of RCC.Methods:1，84paraffin‐embedded specimens,42RCC tissues and corresponding42normal tissues,from the RCC patients were studied with immunohistochemistry (IHC).All specimens were pathologically confirmed.Patients included30men and12women.37cases of Clear cell renal cell carcinoma,3cases of Papillary renal cell carcinoma,2cases of Carcinoma of the collecting ducts.In addition,according to the WHO(1997)histologic Fuhrman grading system and AJCC of TNM(2002) classification system,they were:27cases of T1，9cases of T2，5cases of T3，1case of T4；2，To study the relation of HER2/neu expression between RCC and normal tissue,we divided cases into two groups, RCC group and normal tissues group,and then statistical analysis.In addition,we analyze HER2/neu expression of normal tissues according to TNM stage； 3，6fresh specimens, RCC tissues and normal tissues,obtained in the process of surgical operation were extracted Protein,and then experiment with western blot method.Results:1,Of a total of42RCC tissue and normal tissues respectively, IHC showed that6tumours (14.29%) and28normal tissues(66.66%) over‐expressed the HER2/neu protein；2, The result of statistical about analysis RCC group and normal tissues group, is that the p value=0.007,r=‐0.410.In addition,we analyze HER2/neu expression of normal tissues according to TNM stage. The result is that p=0.027,r=‐0.246；3,For western blot,a total of6specimens from surgical operation，which are divided into6RCC tissues and6normal tissues respectively,2cases of RCC tissue over‐express HER2,however,all normal tissues are positive expression.Conclusions: HER2/neu rarely express in RCC tissues.there are negative correlation between HER2/neu expression of normal tissue and that of RCC, In addition,HER2/neu status of normal tissue are negatively correlated with the depth of TNM stage.
