ObjectiveDetecting the expression of Twist and E-cadherin proteins, to investigatethe role of aberrant expression of the two proteins in colorectal carcinoma.MethodsImmunohistochemistry(IHC) method was used to detect the expression ofTwist and E-cadherin proteins in78cases of colorectal carcinoma,42cases ofadenoma and20cases of paracancerous mucosa tissues; Twist proteinexpression level was detected by Western blot method in30cases ofcolorectal carcinoma and30cases of adenoma.ResultsIHC results:①the positive rates of Twist protein were56.4%(44/78),26.2%(11/42) and10%(2/20), while the positive rates of E-cadherin proteinwere48.7%(38/78),76.2%(32/42) and100%(20/20) respectively in colorectalcarcinoma, adenoma and paracancerous mucosa. The significantdifferences statistically were found between the groups of paracancerousmucosa,adenoma and carcinoma;②Lower positive rates of Twist proteinwere observed in the groups of well-moderate differentiation(40%) andnegative lymph node metastasis(43.2%) than that in the groups of poorlydifferentiation(78.8%) and positive lymph node metastasis(73.5%), significantdifferences statistically were found between the groups. But no significantdifferences was found between the groups of the degree of infiltration; Higherpositive rates of E-cadherin protein were observed in the groups ofwell-moderate differentiation (62.2%), non-lymph node metastasis (59.1%) and early infiltration (74.1%) than that in the groups of poorlydifferentiation(30.3%), lymph node metastasis (35.3%)and advancedinfiltration(35.3%);③The negative correlation between the expression ofTwist and E-cadherin proteins was obtained in colorectal carcinoma)r=-0.488,p=0.000).Western blot results: Higher expression quantities of Twist protein wasobtained in the group of colorectal carcinoma than that in the groups ofadenoma and paracarcinoma mucosa; In the groups of well-moderatedifferentiation and non lymph node metastasis, lower positive rates of Twistprotein was observed than that in the groups of poorly differentiation andlymph node metastasis, significant differences statistically were foundbetween the two groups, but no significant differences was observed betweenthe groups of the degrees of infiltration.ConclusionsIt was suggested that over-expression of Twist protein and low-expressionof E-cadherin protein might play an important role in pathogenesis andprogression of colorectal adenocarcinoma.
