Objective To investigate the effects of telmisartan on the expression of vascular cell adhesmolecule-1(VCAM-1) and the adhesion to monocytes induced by homocysteine(HCY) in human umbilical vein endothelial cells(HUVEC).and to investigate the role of telmisartan on peroxisome proliferator activatedreceptor-delta in the anti-inflammation.Methods Part I Collagenase was used to isolate the endothelial cell from human umbilicvein;Human umbilical vein endothelial cells was identidied byImmunocytochemical stain technique and morphology observation under opticalmicroscope.Part II RT-PCR was used to examine the expression of VCAM-1mRNA.PPAR-δmRNA;WESTERN-BILOTTING was used to detect expression of Pδ protein.; ROSE BENGAL staining techniques was used to detect Mononuclcells-endothelial cell adhesion function.Results(1)We found the cultured cells were oval or polygon and retained cobblestoneappearance through inverted phase contrast microscope. Red fluorescence wasobserved in the cytoplasm under the fluorescence microscope with Rabbit agaihuman Ⅷ factor.(2)The expression of VCAM-1mRNA and the adhesion function to monocytes winduced by HCY in HUVEC comparaed with the control group(P0.01).(3)Comparion with HCY group, telmisartan(10-1000nmol/l)diminished VCmRN Aexpression induced by HCY in HUVEC in a concentration-dependentmanner(P0.01).(4)The expression of PPAR-δmRNA was promoted by telmisartan when HCY iHUVEC comparaed with the control group(P0.01). (5) GW0742obviously decreased the expression of VCAM-1mRNA and theadhesion function to monocytes were induced by HCY in HUVEC(P0.01).(6) GSK0660significantly antagonist the effect of telmisartan decreased theexpression of VCAM-1mRNA and the adhesion function to monocytes wereinduced by HCY in HUVEC(P0.01).Conclusion1High purity HUVECs were obtained by collagenase,which privids an idealexperimental model for pathophysiology studying of vascular endothelial cells ivitro;2Telmisartan decreased the higher expression of VCAM-1mRNA, protein and thlevel of endothelial adhesion with monocytes induced by HCY in HUVEC.Themechanism may be through promote the activation of PPAR-δto controlinflammation and endothelial injury.
