Objective: To investigate the effect of electroacupuncture (EA)pretreatment at the Baihui acupoint of rats induced with central fatigue on thebehaviors,5-HIAA/5-HT and (DOPAC+HVA)/DA ratios in brain, and BUN,CK, LDH, T, SOD, MDA in blood serum of rats.Methods: Male SD rats were randomly divided into six groups:(1) control(2) EA (3)1d fatigue (4)2d fatigue (5) EA+1d fatigue (6) EA+2d fatigue. Wemeasured weight-loaded forced swimming time、spontaneous motor activity andmemory of rats induced with central fatigue after electroacupuncture pretreatmentat the Baihui acupoint. And we measured the5-hydroxyindoleacetic acid(5-HIAA)/5-hydroxytryptamine (5-HT) and [3,4-dihydroxyphenyl-acetic acid(DOPAC)+homovanillic acid (HVA)]/dopamine (DA) ratios in brain of ratsinduced with central fatigue after electroacupuncture pretreatment at the Baihuiacupoint. Measure the level of BUN, CK, LDH, T, SOD and MDA in blood serum of rats.Results: The weight-loaded forced swimming time were not significantdifferences between control and EA(P>0.05). The weight-loaded forcedswimming time of1d fatigue and2d fatigue significantly decreased(P<0.01).The weight-loaded forced swimming time of EA+1d fatigue group was clearlyhigher than that of1d fatigue group(P<0.01), but the weight-loaded forcedswimming time of EA+2d fatigue group and2d fatigue group were notsignificant differences(P>0.05). Spontaneous motor activity were notsignificant differences between control and EA(P>0.05). Spontaneous motoractivity significantly increased in EA+fatigue group than that of fatigue controlgroup(P<0.05). Spontaneous motor activity of1d fatigue and2d fatiguesignificantly decreased(P<0.05). The move time, move distance and movedistance in the center of EA+1d fatigue group was clearly higher than that of1dfatigue group(P<0.01), but the move number were not significant differencesbetween EA+1d fatigue group and1d fatigue group(P>0.05). Spontaneousmotor activity of EA+2d fatigue group and2d fatigue group were not significantdifferences(P>0.05). In the probe trial test, the Percentage of target quadrantand the Number of crossing the platform decreased in1d fatigue group and2dfatigue group than that in control group(P<0.05). The Percentage of targetquadrant and the Number of crossing the platform were not significantdifferences between EA+fatigue groups and fatigue groups(P>0.05).The5-HIAA/5-HT ratio and (DOPAC+HVA)/DA ratio in brain were notsignificant differences between control and EA(P>0.05).5-HIAA/5-HT ratio inmidbrain and striatum and (DOPAC+HVA)/DA ratio in midbrain andhypothalamus significantly inecreased in EA+1d fatigue group than that ofcontrol group(P<0.05).5-HIAA/5-HT ratio and (DOPAC+HVA)/DA ratio in all4regions significantly inecreased in EA+2d fatigue group than that of controlgroup (P<0.05).5-HIAA/5-HT ratio in midbrain and striatum and(DOPAC+HVA)/DA ratio in midbrain significantly decreased in EA+1d fatiguegroup than that of1d fatigue group(P<0.05).5-HIAA/5-HT ratio in all brainregions and (DOPAC+HVA)/DA ratio in hippocampus and hypothalamussignificantly decreased in EA+2d fatigue group compared with that of2d fatiguegroup(P<0.05).The level of BUN, CK, LDH, T, SOD and MDA in blood serum were notsignificant differences between control and EA(P>0.05).(1) The level of BUNof1d fatigue and2d fatigue significantly decreased(P<0.05). The level of BUNof2EA+fatigue groups was clearly higher than that of fatigue groups(P<0.01).(2) The level of CK were not significant differences among6groups(P>0.05).(3) The level of LDH of2d fatigue group significantly increased. The levelof LDH of EA+2d fatigue group was clearly lower than that of2d fatigue group(P<0.01).(4) The level of T of1d fatigue group and2d fatigue groupsignificantly decreased. The level of T of EA+2d fatigue group was clearly higherthan that of2d fatigue group(P<0.01), but the level of T of EA+1d fatiguegroup and1d fatigue group were not significant differences(P>0.05).(5) Thelevel of SOD and MDA were not significant differences between1d fatiguegroup and control group(P>0.05). The level of SOD of2d fatigue significantlydecreased(P<0.05). The level of MDA of2d fatigue significantly increased(P<0.05). The level of SOD and MDA were not significant differences betweenfatigue groups and EA+fatigue groups(P>0.05).Conclusions: The study showed that electroacupuncture pretreatmentincreased weight-loaded forced swimming time and spontaneous motor activity.Electroacupuncture pretreatment decreased5-HIAA/5-HT and (DOPAC+HVA)/DA ratios in some parts of brain of rats induced with centralfatigue. And the study showed that electroacupuncture pretreatment decreased thelevel of BUN and LDH, increased the level of T of rats induced with centralfatigue.
