Objective: To study the relationship between leptomeningeal fibrosis and the PⅠCP of cerebrospinal fluid(CSF) after subarachnoid hemorrhage(SAH) and the effect ofpentoxifylline(PTX) on leptomeningeal fibrosis.Methods: Fifty-four Japanese rabbits were randomized into model group (n=14),blank group (n=10), normal saline group (n=10), experimental group (n=10), controlgroup (n=10). The animal model of rabbits with SAH was made by infusing the autoblood into the cisterna magna twice, but normal saline group was injected normalsaline; Cerebrospinal fluid (CSF) was only pumped from cistema magna the blankgroup. From the second day PTX was injected into the cisterna magna of experimentalgroup on basic of model, and physiological saline group was injected into the cisternamagna of control group on basic of blank group. At the3rdday,7thday,11thday,14thday,21stday,28thday, CSF was collected and the concentrations of PⅠCP in CSF wasmeasured by radi oimmunoassay(RIA) above all groups. At the28thday, all rabbits offive groups were killed to measure and detect the fibrotic degree of leptomeninges byMasson staining.Results: The level of PICP in CSF of model group was obviously higher than thatof blank group and normal saline group(P<0.01); There was no significant differencebetween blank group and physiological saline(P>0.05); The concentration of PⅠCP inCSF of model group rose obviously at the7thday, and kept rising continuously from7thday to21stday, and increased the highest level, and then expressed the high level at28thday; The concentrations of PⅠCP in CSF of experimental group were obviouslydecreasing compared with model group(P<0.05), but were significantly increasingcompared with control group; There was no significant difference between blankgroup and control group(P>0.05). Masson staining: The gray scale of leptomening of model group was obviouslylower than normal saline group and blank group(P<0.05), and the thickness and thecontent of collagen was higher(P<0.05); There was no significant difference betweenblank group and normal saline group in the three respects(P>0.05); The gray scale ofleptomening of experimental group was higher than model group(P<0.05), and thethickness was slightly thinning but no significant difference(P>0.05), and the contentof collagen was obviously lower(P<0.05); Experimental group VS control group: thegray scale of leptomening was obviously decreasing in experimental group(P<0.05),and thickness and the content of collagen was increasing(P<0.05); There was nosignificant difference between blank group and control group in the threerespects(P>0.05).Conclusion:1.The levels of PICP in CSF increased suggest that theleptomeninges were fibrosis after SAH and the high level expressed continuouslyreflect the virtual fibrotic degree;2. PTX can relieve leptomeningeal fibrosis afterSAH and prevent hydrocephalus after SAH.
