Objective:Develop a reliable nonunion model in rats and detect perlecan in the stump of nonunion of bone tissue to explore the role of perlecan in bone nonunion。Methods:Divide sixty-four10-week-old male Wistar rats into2groups randomly. Insert a1.25mm diameter K-wire into the right femoral canal,then a mid-diaphyseal transverse fracture was created by line saws.In the experimental group,the periosteum was then cauterized circumferentially for a distance of2mm on eachside of the fracture.meanwhile, no special treatment was done to normal healing group. After making sure the formation of bone nonunion by general sample observation,histopathological and X-ray examination at the2nd,4th,6th,8th week, we detect the changes of Perlecan in the bone tissues through immune examination.Results:General megascopic sample observation,histopathological and X-ray examination all show that in experimental group, the ends of the fractured bone,filled with fiber scar,become round and were resorbed,with no bone tissue connection at8th week after surgery.Contrarily, the normal group all healed at6th week after fracture. Besides, perlecan was observed along the osteocyte bodies, specially within the peri-and extracellular space.The signal reached a peak at2nd week and decreased with time prolonged.The PN expression of the nonunion fractures was lowerer than that of normal fracture group.Conclusion:The cauterized fracture model could be regarded as a reliable and practical experimental animal model of bone nonunion. Perlecan have relation with the occurrence of nonunion of bone,could promote the heal of fracture and the maturation of bone matrix.
