Objective This study was to observe rats serum which containing the10kinds of Zhuang drug on in vitro human hepatic carcinoma cell SMMC-7721’s effect, screening out drugs with a strong tumor suppressor role. And based on apoptosis and WWOX tumor suppressor gene to discuess its mechanism.Methods Clean-class adult Wistar rats were give10kinds of Zhuang medicine including Jingangteng、Solanum nigrum、detoxification fern、Scutellaria barbata、Paiqian grass、Shijianchuan、the Gynostemma, Touguxiao、Phyllanthus, Actinidiae to prepare drug-containing serum by serum pharmacological method. Use low、middle and high dose group acting on the human hepatoma SMMC-7721cells, set saline (NS) as control group, and cyclophosphamide (CTX) as positive control group. Drug-containing serum act on hepatoma cell measured by MTT assay to detect the cell proliferation at24h and48h, in order to filter out a effective drugs which has tumor suppressor role. Use AnnexinV-FITC/PI dual staining flow cytometry way to detect different concentrations of Jingangteng induced apoptosis rate of human hepatoma SMMC-7721cells. To detect Jingangteng-containing serum affect Hepatoma cell WWOX tumor suppressor gene’s expression by fluorescence quantitative PCR.Results1. In this study, MTT way screening results are shown that the compared with control group, Jingangteng and Scutellaria barbata containing serum can better inhibit in vitro growth of human hepatoma SMMC-7721, and high concentrations of Jingangteng has the strongest inhibitory effect at48h(p<0.05).2. the low and high concentrations of Jingangteng-containing serum act on human hepatoma SMMC-7721cells, the apoptosis rates were30.85%,25.19%. The Jingangteng group apoptosis rate was higher than the blank serum control group. It was56.78%,56.85%of the cyclophosphamide (CTX) positive control group’s apoptosis rate. The Jingangteng group has apparent inhibitory effect on human hepatoma SMMC-7721cells.3. Jingangteng containing serum act on human hepatoma SMMC-7721cells and to detect the WWOX tumor suppressor gene’s expression. The CT value of the low concentration group was33.47±0.80, the high concentration group was34.17±1.01, WWOX gene relative to β-actin gene’s expression were1.05±0.06'0.82±0.04, compared with the blank control group, there were no significant statistical difference (P>0.05).Conclusion Jingangteng-containing serum and Scutellaria barbata containing serum inhibit in vitro the human hepatoma SMMC-7721cell proliferation. Jingangteng-containing serum’s inhibition effect may has something to do with that the Jinggangteng-containing serum promote apoptosis of the human hepatoma SMMC-7721cell. The mechanism has nothing to do with the tumor suppressor gene WWOX’s expression.
