| Objective To observed the expression of visfatin and NF-κB in acute necrotizing pancreatitis(ANP) with kidney injury,and the change of them with the intervention of the low molecular weight heparin(LMWH),to explore the role of visfatin and NF-κB in this pathological process,the effects of LMWH on visfatin and its possible mechanism.Methods A total of96male SD rats were randomly divided into3groups:control group (group C,n=32),ANP group (group A,n=32) and LMWH intervention group (group G,n=32).Rats in group A were narcotized by intra-peritoneal injection of10%chloral hydrate.Then the abdomen median incision was performed,and the pancreatic gland was exposed,bile duct was injected antidromicly by microinjector with1.5%sodium deoxycholate to the establishment of the ANP model.In group C,the rats were underwented operation with nothing infused and the pancreas was flipped and striked gently.Rats in group G were injected with LMWH calcium subcutaneously (10U/100g) after the establishment of ANP model.All rats were sacrificed at6hours,12hours,24hours and48hours after corresponding operation.Then specimens from pancreas and kidney tissues were sliced and stained.To observe the histopathological changes of pancreas and kidney tissues under the light microscope and extract the pathological score at the same time.The level of serum amylase,IL-6,TNF-α was detected;the expressions of visfatin mRNA and NF-κB p65mRNA in kidney tissue were detected by reverse transcription-polymerase chain reaction(RT-PCR);visfatin and NF-κB p65protein in kidey were evaluated by Western Blot.Results The level of serum amylase,TNF-α and IL-6in serum,the pathology score of the pancreas and kidney in group A were evidently higher than those in group C(P<0.05),while in group Qthe pathology score of pancreas and kidney,the level serum amylase,TNF-α,IL-6in serum were significantly lower than group A(P<0.05), but still higher than those group C(P <0.05).In group A,the expression of visfatin mRNA,NF-κB p65mRNA and protein of kidney were significantly higher than those in group C(P<0.05),and those were gradually increased as the time gone on;while in group G changes were lower than group A(P<0.05),but still higher than those in group C(P <0.05).Conclusions (1) The decreased expression of visfatin may be related to the severity of kidney injury in ANP.(2) With ANP occurred,the activation of NF-κB,the massive release of inflammatory factors and the high level expression of visfatin may be lead to kidney injury in ANP.(3) LWMH may alleviate kidney injury in ANP by its anti-inflammatory effects. |
