Effect Of Low Molecular Weight Heparin On The Expression Of P-Selectin And E-Selectin In Acute Necrotizing Pancreatitis With Liver Injury

Objective To investigate the expression of P-selectin and E-selectin in acute necrotizing pancreatitis (ANP) in rats with liver injury and the changes of both of them with the intervention of low molecular weight heparin (LMWH), to investigate the roles of P-selectin and E-selectin in the pathogenesis of ANP, the effects of LMWH on P-selectin and E-selectin and its possible mechanisms.Methods Ninety-six Sprague-Dawley rats were randomly and equally divided into control group(group C, n=32), ANP group(group A, n=32)and LMWH intervention group(group G, n=32). In group A, the rats were induced retrograde injection of1.5%sodium deoxycholate (1ml/kg) into the biliary-pancreatic duct to induce ANP. In group G, after modeling, the rats were given subcutaneous injection of LMWH (10U/100g). In group C, The abdomen was closed only after turning the pancreas of rats.The rats were killed at6h,12h,24h and48h. The pathologic changes of the pancreatic and hepatic tissues were observed and graded under microscope. The level of serum amylase was detected. The level of serum IL-6and TNF-a in serum was determined by ELISA. The expression of P-selectin mRNA and E-selectin mRNA in liver were evaluated by RT-PCR. The protein expression of P-selectin in liver was detected by Western blot.Results The pathologic scores of the pancreatic and hepatic tissues, the level of amylase, TNF-a and IL-6in serum, the expression of P-selectin mRNA, E-selectin mRNA, and P-selectin protein of the liver at6h,12h,24h and48h in group A were significantly higher than those in group C(P<0.05), In group G, the pathologic scores of the pancreatic and hepatic tissues, the level of amylase, TNF-a and IL-6in serum, the expression of P-selectin mRNA, E-selectin mRNA, and P-selectin protein of the liver were decreased significantly compared with the rats in group A (P<0.05). but still higher than those in group C(P<0.05).Conclusions The expression of P-selectin and E-selectin may be related to the severity of ANP with liver injury. LMWH may reduce the lesions of pancreas and liver in ANP by down-regulating the expression of P-selectin and E-selectin.
