Objective:To investigate the expression of Rad50and the histopathological changes in the salivary gland epithelial cells of wistar rats atfer irradiation,discussing the function and significance of Rad50in the repair process of radiation injury of the salivary gland epithelial cells.Methods:36Wistar rats were randomly distributed to6groups,the parotid gland and submandibular gland of each group was irradiated for0,3,6,9,12,15Gy doses of60Co γ-ray. The radiated rats were sacrificed after2hours,and then the parotid glands and submandibular glands were took out for research. The histopathological changes in the parotid gland and submandibular gland of rats were observed with transmission electron microscopy. Expression of Rad50mRNA were evaluated through RT-PCR and expression of Rad50protein were evaluated through Immunohistochemistry method. The experimental data were analyzed for ANOVA with SNK method using SPSS13.0software package.Results:Semiquantitative analysis revealed Rad50mRNA were expressed in all groups. There were no significantly difference of the expression of Rad50mRNA in all groups (P>0.05). Immunohistochemical analysis revealed strong staining for Rad50localized mainly in the serous alveolus and duct of parotid glands in all groups. And it also revealed Rad50localized in the serous alveolus of submandibular glands,but in some groups displayed weak reactivity in mucous alveolus and duct. Expressions of Rad50proteins in the serous alveolus of parotid gland after15Gy irradiation were lower than those in normal,the other expressions of each groups were different but not reached statistical significance.Under the transmission electron microscope, cells of the rat’s salivary glands were injured for dose-dependent.Conclusions:The histopathological changes of Cells in the rat’s salivary glands were injured after2hour high-dose irradiation. It may not influence on the expressions of Rad50by low-dose irradiation.Expressions of Rad50protein in the serous alveolus of parotid gland after15Gy irradiation were lower than those in normal,but other had no statistically different with each other,which suggested that the expression of Rad50in the salivary gland was limited,and perhaps was related to the radiation sensitivity of the salivary glands.
