Objective:To constructe the model of irradiated injury salivary gland and investigate the expression of NBS1genes and proteins in the radiation-injured model of salivary gland in rats, and explore the irradiated injury repair mechanism of NBS1in salivary gland.Methods:The rats were randomly divided into two groups, the irradiated rats(n=40) were fractionated exposed to a dose of3Gy60Co y-rays,given two days time,leading to an calculated dose of3,6,9,12,15Gy; The sham-irradiated controls(n=40) were anesthetized in parallel with the irradiated rats but not irradiated. After irradiation in two to four hours, the rats were sacrificed, IHC and RT-PCR were used to detect the NBS1expression on protein and the mRNA level, the ultrastructural changes were observed by transmission electron microscopy, t-test was used for data analysis.Results:The organization was more atrophy, and the cells were damaged even more serious in parotid gland than in submandibular gland; Compared with the controls, with the groups of dose above9Gy in parotid gland and the dose above12Gy in the submandibular gland, the expression of NBS1mRNA were reduced (P<0.05); Compared with the controls,with the groups of dose above 9Gy in parotid gland and the dose above12Gy in the submandibular gland, the expression of NBS1protein in serous cells, and the dose above12Gy in parotid gland ductal epithelial cells were reduced(P<0.05),and in the ductal epithelial cells as well as mucous cells in the submandibular gland were steadily(P>0.05);Conclusions:The model of irradiated injury salivary gland was successfully constructed; The parotid gland is more radiation sensitive than the submandibular gland, and serous cells is more radiation sensitive than mucous sex cells and ductal epithelial cells.NBS1might be involved in salivary gland irradiated injury repair.
