Objective To construct the rats’ right lower limb with ischemia-reperfusion injury model for three interventions’ protective effection,discuss the effect mechanisms of the methods,to provide experimental evidence and theoretical basis for clinical intervention,prevention and treatment of limb ischemia reperfusion injury.Methods100healthy SD rats were randomly divided into sham operation group, ischemia-reperfusion group, methylprednisolone group, alprostadil group and ischemia post processing group,5groups (n=20).to use the right lower limb ischemia reperfusion model, divided into groups according to the different reperfusion time after ischemia3h,6h,9h,12h, a total of four subgroups, each subgroup of5. Various subjects in the corresponding time points were sacrificed, blood and gastrocnemius tissue, ELISA method for the determination of serum levels of TNF-α, IL-1β of NFκB level, immunohistochemistry, semi-quantitative and homogenate determination of the muscle of MPO, TNF-alpha and MDA content, records muscle wet dry weight ratio, and organization of subtle structural changes in the light microscope.Results Methylprednisolone group, alprostadil group and ischemia post processing group after ischemia reperfusion, serum levels of TNF-alpha, IL-1β of NFκB levels and muscle of MPO, TNF-a and MDA content compared with ischemia reperfusion group in varying degrees,in the light microscope, muscle tiny constitution prompt the extent of skeletal muscle cell edema and inflammatory cell infiltration have been reduced.Conclusions Methylprednisolone, alprostadil, ischemia post processing have protective effection on ischemia-reperfusion injury.
