Effects And Mechanisms Of Seabuckthorn Seed Oil And Seabuckthorn Berry Oil In Rats Model With Ulcerative Colitis

Objective:To study the protective effects of seabuckthorn seed oil and seabuckthorn berry oil on ulcerative colitis, ulcerative colitis model in rats was induced by Dextran sulfate sodium(DSS)in the experiment. To evaluate the efficacy and to explore the anti-inflammatory mechanism of seabuckthorn seed oil and seabuckthorn berry oil treating ulcerative colitis by detecting the levels of TNF-α in the serum, observing the expressions of TNF-α mRNA and nuclear factor-kappa Bp65mRNA(NF-κB p65) and the activity of NF-κB p65in the colon tissues.Method:Thirty-two male rats were randomly divided into four groups:normal control group, DSS model control group, seabuckthorn seed oil treatment group and seabuckthorn berry oil treatment group. Except the normal control group, the other groups were given5%DSS to induce ulcerative colitis model in rats. The rats in the seabuckthorn seed oil treatment group and seabuckthorn berry oil treatment group respectively received a seabuckthorn seed oil and seabuckthorn berry oil treatment daily by gastric lavage, while the remaining two groups received distilled water. The clinical symptom were observed during the experimental period. Three weeks later, all the rats were killed. The macroscopical and histological damages of colon were evaluated. The activity of myeloperoxidase(MPO), superoxide dismutase(SOD)and the content of malondialdehyde(MDA) in the colon tissues and serum were measured by biochemical methods. The serum concentration of TNF-α was measured by using enzyme-linked immunosorbent assay. The expressions of TNF-α mRNA and NF-κB p65mRNA in colonic tissues were measured with the methods of Real-time PCR. And the expression of NF-κB p65in colonic tissues was detected by immunohistochemistry. Results:1.All the rats in the DSS model group can been observed the changes of the symptoms such as intermittent diarrhea with mucus and blood at stool. The scores of DAI was increased obviously. Oedema, hyperemia and anabrosis were observed in the colonic mucosa of experimental colitis. And the UC model of rats was made successfully.2.Compared with the DSS model control group, the scores of DAI were decreased obviously in the seabuckthorn seed oil treatment group and seabuckthorn berry oil treatment group(P<0.01).3.Compared with the DSS model control group, seabuckthorn seed oil and seabuckthorn berry oil notably reduced the macroscopical and histological scores of colon tissues(P<0.01).4.Compared with the DSS model control group, seabuckthorn seed oil enhanced the activity of SOD and reduced the content of MDA in the colon tissues and serum(P<0.05), and reduced the expressin of MPO in the colon tissues(P<0.05). And seabuckthorn berry oil enhanced the activity of SOD in the colon tissues and serum, and reduced the content of MDA in the colon tissues(P<0.05). Compared with the seabuckthorn berry oil treatment group, the activity of SOD in the colon tissues was higher in the seabuckthorn seek oil treatment group(P<0.05).5.Compared with the normal control group, the serum concentration of TNF-a in the DSS model control group was significantly increased(P<0.01), Compared with the DSS model control group,the serum concentration of TNF-α was decreased in the seabuckthorn seed oil treatment group (P<0.01). Compared with the seabuckthorn berry oil treatment group,the serum concentration of TNF-α in the seabuckthorn seed oil treatment group was lower(P<0.01).6.The expression of TNF-α mRNA in the DSS model group was significantly higher than normal control group(P<0.01). Compared with the DSS model group, seabuckthorn seed oil could decreased the expression of TNF-a mRNA(P<0.01). The expression of TNF-a mRNA in the seabuckthorn berry oil treatment group was higher than the eabuckthorn seed oil treatment group(P<0.01). And the significant difference of the expression of NF-κB p65mRNA was not observed during the all groups(P>0.05).7. The expression of NF-κB p65in the DSS model group was significantly higher than normal control group(Pp<0.01). The expression of NF-κB p65in the seabuckthorn seed oil treatment group was lower than the DSS model control group(P<0.01). Compared with the seabuckthorn seed oil treatment group, the protein expression level of NF-κB p65in the seabuckthorn berry oil treatment group(P<0.05).Conclusion:1.Rat model of UC was induced successfully by5%DSS.2.With the administration of seabuckthorn seed oil and seabuckthorn berry oil, the clinical symptom of ulcerative colitis rats was relieved rapidly, the colon injury were alleviated, the ultramicro-pathology damage of colonic mucosa was restored, and the degrees of inflammation were obviously alleviated.3.The different effect between seabuckthorn seed oil and seabuckthorn berry oil may be due to the different content and proportion of the n-3and n-6PUFA.4.The treatment of seabuckthorn seed oil was effective on the UC rats. The antioxidative effect of seabuckthorn seed oil and inhibiting the expression of NF-κB p65may be the anti-inflammatory mechanism of seabuckthorn seed oil.
