Objective:To evaluate the effects of recombinant human growth hormone on neuronal damage after status epilepticus and its possible mechanism, and to provide the possible evidence on animal experiments for clinical treatment of status epilepticus.Methods:54healthy mature male Wistar rats were randomly included into three groups: control group,model group, growth hormone group. Control group included6rats. Every last group (model group and growth hormone group) included24rats (6rats at6hours,12hours,24hours,48hours respectively). The rats of growth hormone group were injected growth hormone (50mg/kg) by intraperitoneal injection before15minutes of making model. The rats of control group and model group were injected the same amount of saline before15minutes of making model. The rats of model group and growth hormone group were injected lithium chloride (127mg/kg) after15minutes by intraperitoneal injection. The rats of model group and growth hormone group were injected fresh pilocarpine (30mg/kg) after18hours. The control group were injected the same amount of saline at the same time points.Then observe carefully and record the performance and time when seizures appears among these rats. Every group rats were killed at the time point of seize end. Observe morphological changes under light microscope, after HE staining.Detect the apoptosis of rat hippocampal neurons using the method of Tunel. Test the expression of Xlap and CasPase-3using immunohistochemical staining. SPSS13.0version was applyed for statistical analysis and P<0.05was used for statistical significance.Results:Positive expression appeared tinyly about CasPase-3,Ⅺap and Tunel in the control group. The positive cells of CasPase-3,Ⅺap and Tunel was observed that increasing appeared after6h of status epilepticus. CasPase-3and Tunel expression top was observed that appeared at24h; Xlap expression top was observed that appeared at12h. CasPase-3and Tunel positive cells in growth hormone group were observed that was distinguishedly degraded compared with model group (except for6h, P<0.05), at every different time points,while the Xlap positive cells was distinguishedly augmented (except for6h, P<0.05).Conclusion:(1)The protein expression of Xlap and CasPase-3changed after status epilepticus, that manifested that the above two protein acted in the neuronal injury after status epilepticus.(2) The pretreatment of growth hormone can relieve neuronal deterioration after status epilepticus by augmenting the expression of Ⅺap and degrading the expression of CasPase-3.(3) Growth hormone have protective action against post-status epilepticus neuronal deterioration in rat through mechanism effect of anti-apoptosis was acting.
