Objective1. To study the expression of the XIAP, caspase-9 and TUNEL-positive cells in Brain ofepilepticus rat;2. To explore the influence and its possible mechanism of the fasudil hydrochloride tohippocampal neuronal of the rats with status epilepticus apoptosis.MethodsWe selected 54 healthy adult male Wistar rats for the following study:1. GroupingAll subjects were randomly divided into three groups: control group(n=6),modelgroup(n=24)and fasudil hydrochloride group(n=24),the latter two groups were divided into foursub-groups:6h,12h,24h,48h, with 6 rats in each sub-group.2. ModelOne hour before modeling: 50mg/kg saline was intraperitoneal injected to the control group andmodel group, the same volume of fasudil hydrochloride was ntraperitoneal injected to fasudilhydrochloride group;1 hour later: 127mg/kg lithium chloride was intraperitoneal injected to the model group andfasudil hydrochloride group;After 18 hours:Preparing the status epilepticus,30mg/kg pilocarpine(freshly prepared) wasintraperitoneal injected to model group and fasudil hydrochloride group,at the same time thesame volume of normal saline was intraperitoneal injected to the control group.3. Experimental methodsObserved and recorded the rats seizures and behavior, after status epilepticus ,the brains wereremoved in the corresponding time points, HE stained the hippocampus, witnessed the changesin morphology and immunohistochemistry with light microscopy,detected the XIAP andcaspase-9 protein expression by SABC,detected the apoptosis of rat hippocampal neurons withTUNEL . ResultsAfter status epilepticus, we got the following results through observation and analysis:1.The expression of XIAP in the hippocampus of model group and fasudil hydrochloride grouphad a significant increase than that in control group, and the peak expression of XIAP appearedin 12h. The number of XIAP positive cells in Fasudil hydrochloride group at each time pointincreased significantly than that in model group (all P <0.05).2. The expression of Caspase-9 in the hippocampus of model group and fasudil hydrochloridegroup had a significant increase than that in control group, and the peak expression of Caspase-9appeared in 24h. The number of XIAP positive cells in Fasudil hydrochloride group at each timepoint decreased significantly than that in model group (all P <0.05).3. The expression of TUNEL in the hippocampus of model group and fasudil hydrochloridegroup had a significant increase than that in control group, and the peak expression of TUNELappeared in 24h. The number of XIAP positive cells in Fasudil hydrochloride group at each timepoint decreased significantly than that in model group (all P <0.05)Conclusion1.After SE, XIAP and Caspase-9 expressed protein were abnormal,from which we can infer thatthe proteins appear in the process after SE.2.By increasing XIAP and inhibiting the expression of caspase-9, fasudil hydrochloride coulddecreased apoptosis,thereby reduced the brain damage caused by status epilepticus.
