Objective:(1) To explore the expressions of β-defensin-2(HBD-2)、β-defensin-3(HBD-3)、 interferon-gamma (IFN-y) in the lesional skin of Psoriasis Vulgaris,non-lesional skin tissues and normal tissues, as well as in mild, moderate and severe groups.(2) Comparison the expression difference of HBD-2, HBD-3, IFN-y between different groups and analysis the correlation of HBD-2and IFN-y, HBD-3and IFN-y.Further discuss the significantce of HBD-2and IFN-y, HBD-3and IFN-y co-expression in psoriasis vulgaris.Methods:RT-PCR (Real-time fluorescent quantitative reverse transcriptate polymerase chain reaction) technique was used to detect the mRNA expressions level of IFN-y, HBD-2and HBD-3with psoriasis lesions samplcs,non-lesional psoriasis skin samples and normal skin samples (45cases of psoriasis vulgaris and15cases normal controls); Skin lesions of psoriasis vulgaris was divided into mild, moderate and severe groups (mild had25cases, moderate had12cases, and severe group had8cases) by PASI. Using RT-PCR method to detect the expressions level of IFN-y mRNA, HBD-2mRNA and HBD-3mRNA in these three groups. Analysis the correlation between these three mRNA expression and PASI in skin lesions,and comparison the difference of HBD-2,3and IFN-y expression level between different groups,then analysis the relationship between HBD-2,3and IFN-y.Results:(1) The value of HBD-2,HBD-3, IFN-y in the skin lesions of patients with psoriasis were significantly higher than that in normal skin of patients with psoriasis or that in health people,and compared to normal control the value of HBD-2,HBD-3, IFN-y in the normal skin of patients with psoriasis were significantly higher, the difference between the comparison was statistically significant (P<0.01);(2) Among severe group, moderate group and minor groups,the value of HBD-2,HBD-3, IFN-y in the severe group was significantly higher than that in the other two groups,and the value of HBD-2,HBD-3, IFN-y in the minor groups were the lowest, the difference between the comparison was statistically significant (P<0.01);(3) There was Significant positive correlated relationship between the level of HBD-2and the level of IFN-γ in all the subjects(Skin lesions of Psoriasis vulgaris group,Non-lesional psoriasis group and the control group),thecorrelation coefficient was0.785(r=0.785).There was also(Significant)positive relationship between the level of HBD-2and the level of IFN-γ in kin lesions of Psoriasis vulgaris group, the Correlation coefficient was0.670(r=0.670);(4) HBD-3and IFN-γ expression levels were positively correlated among all the subjects normal skin tissues and the non-lesion areas of psoriasis and psoriatic lesion areas; HBD-3and IFN-γ expression levels were also positively correlated in psoriatic lesion areas, the Correlation coefficient was0.791and0.662(r=0.791,r=0.662,P<0.05);(5) The expression of HBD-2,-3and IFN-γ had positivecorrelation with PASI in psoriasis lesions(r=0.887,r=0.756,r=0.787,P<0.05).Conclusions: The results of this study show that the expression of HBD-2, HBD-3and IFN-γ inlesions with psoriasis vulgaris were significantly upregulate, the more serious of the lesionswith psoriasis vulgaris,the more stronger of the expression in lesions, The mRNA expression ofHBD-2,-3and IFN-γ had positive correlation with PASI in psoriasis lesions. There is positivecorrelation between IFN-γ and HBD-2, just as IFN-γ and HBD-3.These all show that HBD-2,3and IFN-γ all have closly relations with psoriasis,we reasonably speculate that HBD-2,3andIFN-γ may have interaction, the co-work and overexpression of HBD-2,-3and IFN-γ inpsoriatic lesions may play a key role to contribute to the pathogenesis of psoriasis.
