Objective:The etiology and pathogenesis of psoriasis has not yet fully understood,but recentpapers highlight the role of dysregulated expression of antimicrobial peptides (AMPs) in thepathogenesis of psoriasis. Human defensins (HBD) is one of the important AMPs. Through theinvestigation of the expression of humanβ-defensin -2(HBD-2)、humanβ-defensin -3(HBD-3)and tumour necrosis factor-α(TNF-α) in the lesional and uninvolved skin of psoriasisvulgaris and in normal controls , HBD-2,-3 and TNF-αin the immunological disfunction ofpatients with psoriasis vulgaris was explored.Methods:The skin biopsies were collected from lesional and uninvolved skin of 45 cases of psoriasisvulgaris and 15 cases normal controls. According to PASI, the 45 cases of psoriasis vulgariswere divided into three groups: patients with mild (n=15), moderate (n=15) and severe(n=8).Real-time fluorescent quantitative reverse transcriptase polymerase chain reaction(RT-PCR) was used to examine the mRNA of HBD-2,-3 and TNF-αin the tissue samples.Results:1. The mRNA of HBD-2,-3 and TNF-αwere presented in all specimens from normal control,uninvolved psoriasis and psoriasis lesions.The mRNA expression levels of HBD-2,-3 andTNF-αwere significantly higher in the lesional skin than that in uninvolved psoriasis and normalcontrols, and that in psoriasis lesions were higher than that in uninvolved psoriasis.(P<0.01).2. The mRNA expression of TNF-αhad positive correlation with that of HBD-2,-3in psoriasislesions in psoriasis lesions.(r=0.807、r=0.809, P<0.05).3. The mRNA expression levels of HBD-2,-3 and TNF-αwere significantly higher in severegroup than that in moderate and mild group, and that in moderate group higher were than that inmild group.(P<0.01).4. The mRNA expression of HBD-2,-3 and TNF-αhad positive correlation with PASI inpsoriasis lesions.(r=0.887、r=0.756、r=0.785,P<0.05).Conclusion These findings indicate that overexpression of HBD-2,-3 and TNF-αin psoriaticlesion may contribute to the pathogenesis of psoriasis vulgaris.
