Studies On Extraction Process Of Effective Components Of Semen Ziziphi Spinosae

As a kind of traditional Chinese medicine frequently used, Semen Zizyphi Spinosae is recorded in pharmacopoeia of the people's republic of China (from 1977 to 2000 Edition). Because China has the plentiful resource of Semen Zizyphi Spinosae, it is great significance to study its active components for the effective using of the resource, which can supply scientifical bases and technical supports for the resource exploiture. Total saponins, total flavonoids and total polysaccharides have affinity with the pharmacologic actions of Semen Zizyphi Spinosae. The optimum extractive technology for the three active substances in Semen Zizyphi Spinosae has been studied in the thesis.Because supersonic technique is fast and highly effective for the extraction of effective components in traditional Chines medicines, the extracting and separating the active component in Semen Zizyphi Spinosae by use of the technique is focused attention on in this thesis.First, the extraction procedures of total saponins of Semen Zizyphi Spinosae was studied. The supersonic technique is the best in the method studied. Through the single factor and the three factors (concentration of ethanol, stock ratio, time of extraction) three levels orthogonal test, we confirmed the optimum extractive procedures of total saponins was that the concentration of ethanol was 70%, stock ratio was 1:30, ultrasonic treatment 2 times, and 50 minutes per time. The average extraction ratio was as high as 98.79%.Second, the extraction procedures of total flavonoids of Semen Zizyphi Spinosae was studied. The supersonic technique is the best in the method studied. Through the single factor and the three factors (concentration of ethanol, stock ratio, time of extraction) three levels orthogonal test, we confirmed the optimum extractive procedures of total saponins was that the concentration of ethanol was 70%, stock ratio was 1:30, ultrasonic treatment 2 times, and 50 minutes per time. The average extraction ratio was as high as 98.73%.Last, the extraction procedures of total polysaccharides of Semen Zizyphi Spinosae was studied. The supersonic technique is the best in the method studied. Through the single factor and the three factors (concentration of ethanol, stock ratio, time of extraction) three levels orthogonal test, we confirmed the optimum extractive procedures of total saponins was that the concentration of ethanol was 70%, stock ratio was 1:50, ultrasonic treatment 2 times, and 15 minutes per time. The average extraction ratio was as high as 98.61%.
