| Nitric Oxide Synthase (nNOS)could be got through detecting pariesanterior vaginase from stress urinary incontinence (SUI). This study aimed at exploring the mechanism of nNOS in SUI by analyzing its expression.Methods:Specimens came from anterior vaginase of sixty SUI and fourty-seven NON-SUI patients who carried out operation at the third Hospital ofJi Lin University and Changchun Obstetrics and Gynecology Hospital between2010January and2011Nov.All the specimens were fixed by formalin solution and embedded inliquid paraffin. Streptavidin–peroxidase (S-P) method was used to detectnNOS expression as immunohistochemical staining;and the expression ofnNOS mRNA was mesured by RT-PCR. The datas were analyzed by SPSS18.0software. T-test was used to test measurement data between twoindependent specimens in two normal distribution groups. The personchi-square was used to test enumeration data in these groups. Spearman wasemployed to judge the correlativity between nNOS and SUI. All results from PCR were showed by Mean±SEM and analyzed by ANOVA;Values of P <0.05were thought statistically significant.Results:The expression of nNOS was mainly appreciated in endochylema which distributed generally and showed cinnamomeous or dark brown. In non-menopause group of SUI patients, the nNos positive expression rate was56%in anterior vaginal wall which was higher than10.53%control g roup. In menopause group of SUI patients, the nNos positive expressionrate was65.71%in anterior vaginal wall which was higher than17.86%in control group. Statistical analysis results showed that the expressionof nNOS was significant deviation in non-menopause and menopause groups of SUI patients with control groups(P<0.05); and the expression ofnNOS also has significant deviation in non-menopause and menopause groups of SUI patients(x2=9.45,P<0.05).The result of RT-PCR also demonstrated that the expression of nNOS mRNA at anterior vaginase in POP group was obviously higher than that in control group(P<0.05);and the expression of nNOS mRNA at anterior vaginase in non-menopausewas higher than that in menopause whether in SUI group or in controlgroup(P<0.05).Conclusion:1. The expression of nNos increasing at post-menopause phase mayinduce more NO to be produced which leads to the relaxation of muscles andligaments at the bottom of pelvic floor. At last SUI generates.2. The expression of nNOS is higher in SUI group than that in cancer ofthe cervix group. The reason may be that collagen protein is not apparentlyabnormal in cancer of the cervix group but decreases in SUI group. Thisdecreasing leads to the pedestal relaxation of female urethra and bladder neckand revokes SUI;3. The low expression of nNOS in cancer of the cervix group meansnNOS is independent from it. |
