Bladder cancer is the most common malignant tumor of the urinary system, bladdercancer is the fourth most common malignancy in males, is the eighth largest commonmalignant tumors in female patients. Cystoscopy and cytology are the two most commonmeans of detection.Cystoscopy is invasive and painful,it is difficult to accept and has somecontraindications.Cytology is inexpensive and noninvasive,but its low sensitivity lead tomissed diagnosis of patients,which greatly restricts its clinical applications.FISH test as anemerging means of checks,because of the molecular cytology level of the detection meansand the non-invasive,pain-free advantage is more and more used in clinical works.Objective To discuss the feasibility of the FISH test applied to the auxiliary diagnosisof bladder urothelial carcinoma and effectiveness comparison with urine cytology.Methods In this study, randomly selected85cases of hematuria are urology in-patientsof the China-Japan Union Hospital of Jilin University, with the urine specimens collectedfrom10pm to the second morning.All specimens were detected by FISH and urine cytologytesting.The patients were treated with cystoscopy and biopsy,The tumors were confirmed bypathology after surgery,and were graded according to the2004WHO criteria.In thisstudy,two FISH DNA probe combinations by Beijing GP Medical Technology CO.,Ltd wereused.They are probe combination1(CSP3/CSP7) and probe combination2(GLPp16/CSP17).Each case collected about100ml of urine sample.Through the sample slidepreparation,slide pretreatment,hybridization variability,slides washed, stained,we canobserved the FISH experimental results under a fluorescence microscope,while doing thecell detachment studies.Results The results of the probe GLP p16are negative.The results of the probeCSP3,CSP7,CSP17are amplified diversifications.Amplified amount is positively correlatedwith the pathological grade.The sensitivity of FISH is94.94%and a specificity of100%.Sensitivity of cytology is44.30%and a specificity of83.33%. Conclusion The higher the pathological grading of bladder cancer, FISH-positivemutation is more obvious, the higher the detection rate.FISH examination in the detectionsensitivity is better than cell detachment studies. Specificity level of FISH and cytologyresults are similar.
