Background:The liver is a complex digestive organ in our body, which has a strong regenerative capacity. Liver regeneration after partial hepatectomy that is dependent on the regenerative capacity of the residual liver tissue. Recently, the mechanism of liver regeneration after injury which regulated by traditional Chinese medicine has become a hot spot. This experiment was based on this design.Purpose:To investigate the effect of Sinomenine on liver regeneration after partial hepatectomy and its kidney toxicity.Methods:40clean level SD rats, male and female are half, random into normal group (10), control group (10) and the treatment group (20). Normal rats without treatment, control group and the treatment group, will do left lobe grafts resection, and establish liver damage model. The treatment group:Sinomenine to40mg/kg iv, preoperative24hours,1hour after operation. The control group:to physiological saline40mg/kg iv, preoperative24hours, after1hour each. The1,2,3,5,7day after operation get materials, take liver right leaf organization near surgery section3*3*3cm in size, remove the left kidney, pick the eye to take blood, blood serum frozen spare a separation detection ALT, AST, BUN, CREA. Specimens to4%neutral formaldehyde save, HE dyeing read piece, any liver tissue immunohistochemical, detection TGF-β1, VEGF expression.Results:The index of liver regeneration in study group was higher compared with control group (P<0.05). The levels of ALT and AST decreased more significantly in the treatment group than in the control (P<0.05), and the values of BUN and CREA were similar in the control and treatment group (P>0.05). The result of HE staining showed that the effect of liver regeneration in treatment was more effective than that of control. No significant difference was found in kidney toxicity between normal group and treatment group. The result of immunohistochemical implied that the expression of TGF-β1in experimental group was lower than the control group, while VEGF expression was no significant difference between two groups.Conclusion:The effect of Sinomenine on liver regeneration was mediated by inhibiting TGF-β1expression in the liver tissue, and it is no significant correlation with VEGF expression. Sinamenine showed a few of toxicity to kidney to rats.
