Objective:To investigate the expression of Notch1, Jagged1,p-Akt and NF-κB in gastric cancer tissues and their correlation with clinicopathological features and Helicobacter pylori(H.pylori) infection in order to further explore the possible mechanism of gastric carcinogenesis.Methods:60cases of gastric cancer tissues and corresponding adjacent tissues were identified as research objects and20cases of normal gastric tissues were chosen as control.The protein expressions of Notch1,Jaggedl,p-Akt and NF-κB in60cases of gastric cancer tissues,60cases of adjacent gastric cancer tissues and20cases of normal gastric tissues were detected by immunohistochemistry staining.H.pylori infection was tested by Warthin-Starry silver stain.Results:1.The positive rates of Notch1in gastric cancer,adjacent gastric cancer and normal gastric mucosa tissues were40.0%,81.7%,85.0%respectively.There were significant difference among the three groups(x2=27.046,P=0.000) and between the gastric cancer and adjacent gastric cancer group(χ2=21.860,P=0.000) and between the gastric cancer and normal gastric mucosa group(χ2=12.158,P=0.000), there was not significant difference between the adjacent gastric cancer and normal gastric mucosa group(x2=0.000,P=1.000).2.The positive rates of Jaggedl in gastric cancer,adjacent gastric cancer and normal gastric mucosa tissues were70.0%,96.7%,100.0%respectively.There were significant difference among the three groups(x2=21.311,P=0.000) and between the gastric cancer and adjacent gastric cancer group(χ2=15.360,P=0.000) and between the gastric cancer group and normal gastric mucosa group(χ2=6.117,P=0.013),there was not significant difference between the adjacent gastric cancer group and normal gastric mucosa group(P=1.000).3.The positive rates of p-Akt in gastric cancer,adjacent gastric cancer and normal gastric mucosa tissues were55.0%,30.0%,5.0%respectively.There were significant difference among the three groups(χ2=18.357,P=0.000) and between the gastric cancer and adjacent gastric cancer group(χ2=7.673,P=0.006) and between the gastric cancer and normal gastric mucosa group(χ2=15.345,P=0.000), there was not significant difference between the adjacent gastric cancer group and normal gastric mucosa group(χ2=3.888,P=0.049).The positive rates of p-Akt in high/medium differentiation and low differentiation group were21.1%and70.7%,there was significant difference between the two groups(χ2=12.947,P=0.000). The positive rates of p-Akt in T1/T2and T3/T4group were20.0%and62.0%,there was significant difference between the two groups(χ2=4.364,P=0.037).4.The positive rates of NF-κB in gastric cancer,adjacent gastric cancer and normal gastric mucosa tissues were66.7%,36.7%,0.0%respectively.There were significant difference among the three groups(χ2=29.490,P=0.000) and between the gastric cancer group and adjacent gastric cancer group(χ2=10.812,P=0.001),and between the gastric cancer group and normal gastric mucosa group(χ2=26.667,P=0.000) and between the adjacent gastric cancer group and normal gastric mucosa group(χ2=10.115,P=0.001).The positive rates of NF-κB in high/medium differentiation group and low differentiation were42.1%and78.0%, there was significant difference between the two groups(χ2=7.548,P=0.006).The positive rates of NF-κB in T1/T2and T3/T4group were30.0%and74.0%,there was significant difference between the two groups(χ2=5.415,P=0.020).5.Positive correlation was shown between Notch1and Jagged1(r=0.460,P=0.000) or p-Akt and NF-κB(r=0.284,P=0.028).Negative correlation was shown between Notch1and p-Akt(r=-0.287,P=0.026) or Notch1and NF-κB(r=-0.361,P=0.005).No correlation was shown between the expressions of them and H.pylori infection(r=-0.032,r=0.155,r=0.055,r=0.028,P>0.05).Conclusion:1. There were low expressions of Notch1and Jagged1and high expressions of p-Akt and NF-κB in gastric cancer tissues,and there was significant difference between the abnormal expressions of p-Akt and NF-κB and the differentiation status and depth of invasion.These indicated that Notch1,Jagged1,p-Akt and NF-κB played an important role in promoting the development of gastric cancer.2. Positive correlation was shown between Notch1and Jagged1or p-Akt and NF-κB,and negative correlation was shown between Notch1and p-Akt or Notch1and NF-κB in gastric cancer tissues.These indicated that the low expression of Notch1and Jagged1may promote the gastric carcinogenesis though inducing the high expression of p-Akt and NF-κB.3. The positive rate of H.pylori in gastric cancer was lower than adjacent gastric cancer,and its positive rates in gastric cancer and adjacent gastric cancer tissues were no association with the expressions of Notch1,Jagged1,p-Akt and NF-κB.
