Objective:To investigate the expression of Notch1,Jagged1,COX-2and Snail in gastric cancer as well as their correlations with the clinical pathology and helicobacter pylori infection.Methods:The expression of Notch1, Jagged1,COX-2and Snail proteins were detected in60cases of gastric carcinoma,60cases of normal gastric mucosa tissue and20cases of normal gastric tissue by tissue micmarray technique and immunohistochemical staining. For detection of H.pylori infection,Warhin-starry staining was used.Results:(1)The positive expression rates of Notchl,Jagged1proteins in gastric cancer tissue(40.0%,70.0%)were significantly lower than that normal gastric mucosa(81.7%,96.7%)and normal gastric tissue(85.0%,100.0%),there was statistical significance(P<0.05),but was not significant between normal gastric mucosa and normal gastric tissue(P>0.05).The positive expression rates of COX-2and Snail proteins in gastric cancer (60.0%,53.3%) tissue were significantly higher than that in normal gastric mucosa(35.0%,26.7%) and normal gastric tissue(0.0%,5.0%),respectively,there was statistical significance(P<0.05).(2)The expression of Notchl,Jagged1proteins were not correlated with Clinic Pathologic Feature (P>0.05).The expression of COX-2and Snail proteins were closely correlated with the pathologic differentiation degree (χ2=6.213,P=0.013;χ2=8.155,P=0.004)and depth of invasion (χ2=6.125,P=0.013; χ2=3.871,P=0.049), but not correlated with sex, age, the size of tumor and lymph node metastasis (p>0.05).(3)Notchl protein expression was correlated with Jagged1protein expression in gastric cancer (r=0.460,p=0.000);The expression of Jagged1was no relationship with COX-2,Snail expression(r=-0.175,-0.16,bothP<0.05);The level of Notchl expression was negatively correlated with COX-2,Snail expression(r=-0.314,-0.262,bothP<0.05);COX-2protein expression was correlated with Snail protein expression(r=0.357,P=0.005).(4)The infection rate of H.pylori in gastric cancer76.6%(46/60) were lower than that in normal gastric mucosa91.7%(55/60),there was statistical significance (χ2=6.536,P=0.011).The expression of Notch1,Jagged1,COX-2and Snail was no correlation with the infection of H.pylori(r=0.032,0.155,-0.129,-0.042;allP<0.05).Conclusion:(1)The low expression rates of Notchl and Jaggedl proteins in gastric cancer tissue,COX-2and Snail proteins the high expression rates in gastric cancer tissue.The expression of Notchl,Jaggedl proteins were not correlated with Clinic Pathologic Feature,The expression of COX-2and Snail proteins were closely correlated with the pathologic differentiation degree and depth of invasion.But they are not correlated with H.pylori infection.(2)Notchl,Jagged1, COX-2,Snail and H.pylori may be involved in the tumorigenesis and development of gastric.The expression of Notch1and Jagged1were down-regulation that may be promote tumorigenesis of gastric through the up-regulation of COX-2, Snail expression;H.pylori infection may be plays an important role at the early stage of development of gastric cancer.
