Fingerprint refers to some Chinese herbal medicine or traditional Chinese medicine preparations afterappropriate treatment,the use of certain analytical tools get marked its chemical characteristics of thechromatogram or spectra.It can reflect more fully the type and quantity of the chemical composition oftraditional Chinese medicine and its preparation contained,and thus an overall description and evaluation ofthe quality of pharmaceuticals.On the base of the total review of chemical components and pharmacology of Eucommia leaves andmale flowers,a HPLC fingerprint technology is used to determinate contents of three kinds of mainsecondary metabolites geniposides acid、chlorogenic acid、geniposides in Eucommia staminiferous plant、Eucommia male plant、 Eucommia male flower on the determination and analysis of componentaccumulation, it is necessary to provide the reference materials for Eucommia staminiferous plant、Eucommia male plant、Eucommia male flower about quality control and industrialization.Meanwhile,thisarticle also analyzes the chemical composition and pharmocology,and rrfluxing of the acid and alcoholextracted from the honeysuckle green leaves chlorogenic acid,macroporous resin and ethyl acetate werepurified to study to obtain high purity of chlorogenic and extraction and purification of chlorogenic acid inindustrial production to provide detailed information.This study results are as follows:(1)Established the HPLC fingerprint about the green leaves of Eucommia staminiferous plantand Eucommia male plant.Chromatographic conditions is that:Ameritech column(250mm×4.6mm,5μm),the mobile phase was methanlo with0.05%phosphoric acid in the gradient elution mode.The temperatureof column was maintained at25℃.The flow rate was1.0mL/min.The detective wavelength was set at254nm.The detective time was60min. Evaluating the green leaves of Eucommia staminiferous plant andEucommia male plant to generate the control fingerprint under these conditions.Identifiled19commoncharacteristics of the peak and the main secondary metabolites of geniposide acid、chlorogenic acid、geniposide content determination.Derived the high similarity fingerprints about Eucommia staminiferous plant and Eucommia male plant,which contain similar chemical composition,but the specific contentsare different.(2)Established the HPLC fingerprint about the Eucommia male flowers.Chromatographic conditionsis that:Ameritech column(250mm×4.6mm,5μm);the mobile phase was methanlo with0.05%phosphoricacid in the gradient elution mode.The temperature of column was maintained at25℃.The flow rate was1.0mL/min.The detective wavelength was set at254nm.The detective time was70min. Evaluating thedifferent date the fingerprints of male flowers to generate the conrrol fingerprint identified22commoncharacteristics of the peak, and research of secondary metabolites in geniposide acid、chlorogenic acid、geniposide content accumulation,and the picking time of Eucommia ulmoides male flower in full bloomwould be recognized as the opimal one.(3)Established the HPLC fingerprint about the honeysuckle flower buds,stems、green leaves、fallenleaves to evaluate the chlorogenic acid.chromatographic conditions is that:column:Thermo Syncroniscolumn(250mm×4.6mm,5μm);the mobile phase was methanlo with0.05%phosphoric acid.Thetemperature of column was maintained at25℃.The flow rate was1.0mL/min.The detective wavelengthwas set at327nm.The order of chlorogenic acid content is:bud>green leaves>stems>fallen leaves.(4)With acid alcohol reflux method to determine chlorogenic acid optimum extraction process:12times the amount of80%ethanol,ph=5,reflux,each2.5h for the optimum extraction conditions.The bestmacroporous resin medel is HPD100and HPD826,purified extract2macroporous resin.Then6~8timeswith ph=3ethyl acetate extraction, concentration,recrystallization, and get apurity of74.8%chlorogenicacid.
