Objective:The aim of this study was to investigate the anticancer effect of3-bromopyruvate (3-BrPA) on human HeLa cells, and chemosensitivity and radiosensitivity effects in HeLa cells.Methods:1The effect of3-BrPA on HeLa cells proliferation and chemotherapeutic sensitivity were analyzed via MTT assay.2Hoechst33258staining assay was used to observe3-BrPA-induced morphological changes by fluorescence microscopy.3The flow cytometry (FCM) was used to analyze the distribution of cell cycle and the mortality of HeLa cells.4Cell survival of3-BrPA combined with various X-rays was evaluated by colony formation assay and determine the radiosensitivity effect of3-BrPA on HeLa cell.Result:1Treated with3-BrPA at different concentrations (50、100、150、200、250μmol/L) for24h and at different times (2、4、6、24h) for200μmol/L3-BrPA, there were obvious growth inhibition of HeLa cells in does-nd time-dependent manners (P<0.01). Combined with low does of3-BrPA(25μmol/L) for72h, the inhibition rate of cisplatin(DDP,5,10mg/L) on HeLa cell proliferation were increased from (36.6±2.3)%nd (76.5±2.1)%to (84.5±0.9)%nd (94.7±2.5)%espectively(both, P<0.01), while the inhibition rate of3-BrPA (25μmol/L) on HeLa cell proliferation was only (16.7±3.4)%.2Hoechst33258staining showed that cell nucleus concentrated and apoptotic bodies. 3FCM analysis suggests that3-BrPA could induce G2/M phase arrest and lead to apoptosis and necrosis on HeLa cells.4The average deadly dosage(Do) of HeLa cells treated by medication decrease(1.64Gy、2.18Gy), the Quasi-domain dose(Dq) value is less than that in the sole irradiation group(0.78Gy、1.44Gy). Sensitization enhancement ratio (SER) is1.34.Conclusions:The result in our study suggested that3-BrPA could inhibit the proliferation, enhance chemosensitization and radiosensitization, and induce apoptosis and necrosis and G2/M phase arrest of HeLa cells in vitro. In our present study, we have confirmed the antumor activities of3-BrPA, and this provides an experimental basis for the further analysis of3-BrPA.
