Objective:This study is aimed to investigate the molecular mechanisms of LDI on bone marrow-derived endothelial progenitor cell.Methods:take femur and tibia of Sprague-Dawley rats, then repeatedly flushing the bone marrow cavity, after centrifugation seeded in25cm2culture flasks at37℃,5%. The cells were identified by flow cvtometry. the experimental group is given25mGy,50mGy,75mGy100mGy,1Gy X-ray irradiation while the control group not irradiated. Calculate the number of cells at6h、1d、3d、5d、7d after irradiation. cells proliferation was detected by CCK-8. Western-blot were employed to quantify the expression of VEGFR-2; Reverse transcription-polymerase chain reaction (RT-PCR) were employed to quantify the expression of VEGFR-2. Through using immunohistochemisty for observation of the expression of VWF, VEGF receptor2(VEGFR-2/flk-1).Results:after low-does irradiation, the cells count and cells proliferation of experimental group has significantly enhanced compared to control group (P<0.05), While in the1000mGy group, which was set as high dose irradiation, the proliferation of EPCs was decreased, the serum VEGFR-2in the LDI groups were higher than those of control groups by western-blot (P<0.05), Elevated mRNA expression of VEGFR-2were seen at all time points in the LDI group compared with that of control group (P<0.05). Immunohistochemistry of cells revealed stronger expression of VWF, VEGFR2, in the LDI group compared with that of control group (P<0.05).Conclusion:The results indicate that Low-does X-ray irradiation have excited effection on endothelial progenitor cells; Low-does X-ray irradiation also can promote the cells proliferation and cell markers expression of bone marrow-derived endothelial progenitor cells.
