Objective:Explore the local Low Dose X-ray Irradiation(Low- dosed Irradiation, LDI) the impact on the neovascularization of ischemic skin flap of rats and the mechanism.Methods:Between December 2014 and June 2015 selected 100 Sprague wrote two months-Dawley rats, were randomly divided into experimental group(group A) and control group(B), each group of 50. In each rat’s back in the middle of design 1 9 cm x 3 cm ischemia random flap, department near the end, then place after dealing with the disinfection of biological isolation membrane under the flap, then put the flap suture in situ. Flap suture group A rat after the back flap an immediate low dose X-ray single local irradiation, total dose value of 0.2 Gy; Group B at postoperative irradiation did not receive. In postoperative1 d, 3 d, 5 d, 7 d and 14 d respectively on two groups of flap survival situation line gross observation and calculate the flap survival rate; In turn cut specimens in microscope and laser confocal microscope its angiogenesis, by measuring students to calculate the content of intravascular CD105 capillaries.Results:Experimental group A in postoperative 3 d, 5 d, 7 d and 14 d, the survival rate of skin flap, respectively(65.94 + 3.92) %,(45.01 + 4.02) %,(31.87 + 4.19) %, were higher than in control group B(42.83 + 4.88) %,(28.08 + 2.89) %,(15.11 + 4.94) %.Respectively compared to the experimental group and control group to observe the postoperative 1, 3, 5, 7, 14 days flap specimens of fluorescent dye section found: in the experimental group flap endothelial activation increased obviously, new blood vessels form significantly more than the control group, experimental group arrangement of new blood vessels within the lumen is neat, many layers of blood vessels, lumen complete and rules; The control group, less visible endothelial cell activation and lumen arrangement of new blood vessels is a bit poor, less layers of early vascular, and visible tissue edema is obvious.Postoperative 1, 3, 5, 7, 14 days, the experimental group flap NMVD value respectively(28.27 + 2.96),(21.41-2.41),(15.24-1.81),(20.38-2.55) and(30.28 + 3.66)(article/mm squared); Were significantly higher than that of control group flap NMVD(14.94 + /- 2.01),(9.13-1.03)(6.92 + 1.07)(10.07 + /- 1.81) and(18.21 + 2.11)(article/mm squared).Conclusion:Low dose X-ray irradiation can improve the survival rate of ischemic skin flap. The possible mechanism of low dose of X-ray irradiation can promote the activation of endothelial cells, increase the expression of VEGF, by promoting angiogenesis within the skin flap, increase the ischemic flap survival rate.
