Objective: The study of stem cells to treat end-stage cardiovascular disease, has beengradually carried out in many countries of the world, and has become a hot research field.Large and medium-sized hospitals of china have carry out the basic and clinical research, studies haveshown that biological treatment with stem cells transplantation led to improve severe, especially thelife quality of patients with end-stage cardiovascular disease. Based on such an idea as discussthe issue of bone mesenchymal stem cells transplantation for the treatment of valvulardisease of heart failure, the study start from establish the valve disease in rats with heartfailure model and control the heart failure indicator changes of model rats before and afterbone marrow mesenchymal stem cell transplantation, Investigate the method of valvularheart disease heart failure rat model, and vivo experimental studies of bone mesenchymal stemcell transplantation for treatment of rat valvular disease of heart failure the safety and therapeuticeffects.Methods: Use male Wistar rats (about350g,11weeks old) were20consecutive dailysubcutaneous injection of5-hydroxytryptamine (20mg/kg) to establish the heart failuremodel of valvular heart disease. Take the6-8w age Wistar rat femur bone marrow, culturedto the third-generation, line vivo transplantation to valvular disease of heart failure ratmodel. The vivo experiments were divided into three groups: Group A (control group), Bytail vein injection of the DMEM medium1ml; Group B, By tail vein injection of the bonemesenchymal stem cells culture supernatant1ml; Group C, By tail vein injection of thethird generation of1×10E7bone mesenchymal stem cells. Observe homing BMSCstransplantation by small animal in vivo imaging techniques. By echocardiography, BNPand other check, comparison the heart failure indicators before and after transplantationResults: In the modeling process, a rat in the administration of the late died of heart failure. Rats continuous subcutaneous administration of8weeks, echocardiography,pathology confirmed that the whole group of rats has emerged because of univalve orcombined valve disease caused by heart failure. By intravenous injection of the target doseof graft, not found in rats with acute deaths caused by transplantation, not found theobvious complications of organ or systems either. Between the two groups control andself-control, found that after transplantation group B and group C rats are batter than groupA in survival, survival rate, EF, BNP and so on, The heart failure caused by valvular heartdisease were corrected to a certain extent.Conclusions: Use5-hydroxytryptamine continuous administration to establish Wistarrats with valvular heart disease heart failure model is feasible and effective. The samekinds of bone marrow mesenchymal stem cell culture supernatant and BMSCstransplantation is safe in Wistar rats, the transplantation played a certain therapeutic effecton a rat model, consider the possible mechanism related to the bone mesenchymal stemcells in paracrine and differentiation...
