Objective:The Minor Purgative Decoction is composed of Rhubarb, Magnolia and Citrus aurantium, the content of anthraquinones and tannins can affect the efficacy of the Minor Purgative Decoction. The subject explore the efficacy of the material and determine the best decocting time of the rhubarb by determination of content combined with the pharmacodynamics inspection, sothat we can provide the basis for clinical applications of the Minor Purgative Decoction.Methods:As the anthraquinones, tannin, magnolol, honokiol, hesperidin an indicator, to determine the content of total anthraquinone and tannin by ultraviolet spectrophotometry, to determine the content of free anthraquinone and combined anthraquinone, magnolol, honokiol and hesperidin by High Performance Liquid Chromatography, and combined with a fingerprint comparison, to investigate the influence of different decocting time on the content of chemical composition in the Minor Purgative Decoction, clear the efficacy of pharmacodynamic by different decocting time of the rhubarb, explore the pharmacodynamic material basis of the Minor Purgative Decoction, in order to determine the best decocting time of the rhubarb.Results:1、The results of the effect on the content of total anthraquinone in the Minor Purgative Decoction by different decocting time of the rhubarb: The results show that within60min, the content of total anthraquinone gradually increased with the decocting time of the Rhubarb, the highest content shows at45min, and then gradually decreased2、The results of the effect on the content of free anthraquinone and combined anthraquinone in the Minor Purgative Decoction by different decocting time of the rhubarb:The results show that within60min, the content of free anthraquinone gradually increased with the decocting time of the rhubarb, the highest content shows at45min, and then gradually decreased; the content of combined anthraquinone gradually increased with the decocting time of the rhubarb, the highest content shows at30min, and then gradually decreased.3、The results of the effect on the content of Tannin in the Minor Purgative Decoction by different decocting time of the rhubarb:The results show that within60min, the content of tannin gradually increased with the decocting time of the rhubarb, gradually stable at45min.4、The results of the effect on the content of Magnolol and Honokiol in the Minor Purgative Decoction by different decocting time of the rhubarb:The results show that within60min, the content of Magnolol and Honokiol gradually decreased with the decocting time of the rhubarb.5、The results of the effect on the content of Hesperidin in the Minor Purgative Decoction by different decocting time of the rhubarb:The results show that within60min, the content of hesperidin gradually decreased with the decocting time of the rhubarb.6、The results of the TLC fingerprinting on the Minor Purgative Decoction of different decocting time of the rhubarb:The results show:there are irregular changes in the chemical composition on the process of Rhubarb decoction of Rhubarb、Magnolia officianlis、Citrus aurantium:Some showed a growing trend, and some decreased, some irregular change7、The results of the HPLC fingerprinting on the Minor Purgative Decoction of different decocting time of the rhubarb:The results show:Within60min, there are irregular changes in the chemical composition on the process of rhubarb decoction of the Minor Purgative Decoction, especially at30min, there are a large number of new chemical compounds been produced, and then decompose gradually disappear. 8、Pharmacodynamics research results:The results show:The different decocting time of the Minor Purgative Decoction all can significantly promote the end of the normal mouse small intestine carbon advance rate, can increase the volume of the mouse’s small and large bowel, can significantly promote constipation mouse small intestine carbon propelling rate, can shorten the the defecation incubation period and increased defecation grains and also can significantly enhance the ability of movement of the isolated rabbit bowel. The effect intensity:the decocting time of the rhubarb in the Minor Purgative Decoction:10min>20min>30min>45min>5min.Conclusion:The different decocting time of the Minor Purgative Decoction has a profound effect on the chemical composition and pharmacodynamics, the results showed that the efficacy of the Minor Purgative Decoction for a variety of combined effects of substances, to ensure the stability of the efficacy of the Minor Purgative Decoctiond and protect the best clinical drug effects, the best decocting time of the rhubarb in the Minor Purgative Decoction is fixed on10~20minutes.
