Rhubarb is dried roots and rhizomes of Polygonaceae Rheum palmatum L.or Rheum tanguticum Maxim.ex Balf.or Rheum officinale Baill.,for 2015 edition of Chinese Pharmacopoeia included medicinal herbs.It has diarrhea attack plot,clearing heat and detoxicating,removing blood stasis dredging,dampness resolving effect is common traditional Chinese medicine.In the prescription of rhubarb,the purging effect is commonly used.Modern research prove that rhubarb play purging effect is the rhubarb anthraquinone components,including aloe emodin,rhein,emodin,chrysophanol and emodin ether,anthraquinone components in rhubarb exist with two forms: free anthraquinone(aglycone)and the combined anthraquinone(glycosides),and anthraquinone for the corresponding free anthraquinones with sugar formation.The main mechanism is that oral combined anthraquinone to reach the colon,is the enzyme solution for free anthraquinone to play the role of diarrhea,if directing oral free anthraquinone will be absorpted by the digestive tract,the effect will be very weak.The results of study showed that rhubarb had almost no effect if oral free anthraquinone,but had produced the corresponding effect if colon specific drug delivery technology was used to located in the colon to release.Rhubarb free anthraquinone made of the oral colon positioning release particles,compared with rhubarb,to achieve the purpose of synergism.The 2015 edition of Chinese Pharmacopoeia in the quality standard of rhubarb preparations did not make a request for the content of the combined anthraquinone,however,the content of combined anthraquinone can lead to a significant difference in the effect of diarrhea.In this study,the free anthraquinone,combined anthraquinone and total anthraquinone of aloe emodin,rhein,emodin,chrysophanol and physcion was simultaneous measured by HPLC method in rhubarb preparations(Qinghuotablets,Sanhuang tablets and Yiqing granules).With the Ward minimum variance square sum to clustering analysis aloe emodin,rhein,emodin,chrysophanol and physcion of free anthraquinone and combined anthraquinone in rhubarb preparations(Qinghuo tablets,Sanhuang tablets and Yiqing granules)assays from different manufacturers and different batches of the same company.This work is significance for improving control of quality and better control of the efficacy of the rhubarb has guiding significance.Objective:comparing and clustering analysis the contents of aloe emodin,rhein,emodin,rhubarb phenol and physcion of free and combined anthraquinone assays from Commercially available different manufacturers rhubarb preparations(Qinghuo tablets,Sanhuang tablets and Yiqing granules)and carries on cluster analysis.Study on the quality control of the rhubarb preparations on the effect of diarrhea.Methods:1.Established methods for determination the content of rhubarb preparations(Qinghuo tablets,Sanhuang tablets and Yiqing granules)by HPLC.2.Determined the free anthraquinone,combined anthraquinone and total anthraquinone of aloe emodin,rhein,emodin,chrysophanol and physcion was simultaneous measured by HPLC method in rhubarb preparations(Qinghuo tablets,Sanhuang tablets and Yiqing granules).3.Analysed the free anthraquinone and combined anthraquinone of aloe emodin,rhein,emodin,chrysophanol and physcion with ward method by cluster analysis.Results:1.The result of methodology examination establishes showed that the method of HPLC in vitro analysis is a scientific and reliable quality evaluation method.2.The contents of free anthraquinone assays from ten manufacturers Qinghuo tablets aloe emodin,rhein,emodin,rhubarb phenol and physcionwere6.178,1.659,4.479,0.828,2.971,1.949,3.022,5.727,1.933,3.132 mg/g and the contents of combined anthraquinone assays from ten manufacturers Sanhuang tablets aloe emodin,rhein,emodin,rhubarb phenol and physcion were2.282,0.468,0.429,0.496,2.404,1.055,0.304,1.424,2.446,0.830mg/g;The contents of free anthraquinone assays from ten manufacturers Sanhuang tablets aloe emodin,rhein,emodin,rhubarb phenol and physcion were6.272,9.143,3.950,6.718,7.214,5.411,8.501,5.411,11.725,8.785mg/g and the contents of combined anthraquinone assays from ten manufacturers Sanhuang tablets aloe emodin,rhein,emodin,rhubarb phenol and physcion were 3.781,0.700,6.060,4.202,5.560,8.369,6.036,5.621,1.624,3.361 mg/g.The contents of free anthraquinone assays from a manufacturer Sanhuang tablets aloe emodin,rhein,emodin,rhubarb phenol and physcion were 8.785,8.749,9.226,13.884,7.411,6.756 mg/g,the contents of combined anthraquinone assays from a manufacturer Sanhuang tablets aloe emodin,rhein,emodin,rhubarb phenol and physcion were3.361,7.008,3.322,3.345,3.682,4.429 mg/g;The contents of free anthraquinone assays from ten manufacturers Yiqing granules aloe emodin,rhein,emodin,rhubarb phenol and physcion were 0.082,0.108,0.623,0.270,0.390,0.209,0.612,0.052,0.565,0.107 mg/g and the contents of combined anthraquinone assays from ten manufacturers Yiqing granules aloe emodin,rhein,emodin,rhubarb phenol and physcion were 0.175,0.094,0.257,0.333,0.142,0.454,0.501,0.063,0.262,0.110 mg/g.The contents of free anthraquinone assays from a manufacturer Yiqing granules aloe emodin,rhein,emodin,rhubarb phenol and physcion were 0.209,0.193,0.260,0.357,0.241,0.261 mg/g,the contents of combined anthraquinone assays from a manufacturer Yiqing granules aloe emodin,rhein,emodin,rhubarb phenol and physcion were0.454,0.416,0.473,0.300,0.395,0.339 mg/g.3.The results of cluster analysis showed that the outstanding characteristics of the category division,free and combined anthraquinones content has the spatial distribution characteristics in different manufacturers,contents of combined anthraquinone and free anthraquinone of aloe emodin,rhein,emodin,chrysophanol and physcion assays from different manufacturers in rhubarb preparations(Qinghuo tablets,Sanhuang tablets and Yiqing granules)was significantly different.Conclusions:The result of methodology examination establishes showed that the method of HPLC in vitro analysis is scientific and reliable.The contents of total anthraquinone which as major components of rhubarbvaries assays from commercially available ten manufacturers in rhubarb preparations(Qinghuo tablets,Sanhuang tablets and Yiqing granules)is difference,especially the contents of combined anthraquinone which plays as purging constitution is different as well.
