ObjectiveThe research was established on the foundation that kidney-yang asthenia syndrome exist the functional change of HPG axis, to observe that dismantled shenqi pill groups how to influence kidney-yang asthenia syndrome animals’ body weight, organ weight index, rectal temperature, spontaneous activity, antifatigue and hypoxia tolerance test, HE pathological dyeing, and the content of PRL, FSH, LH, E2, T, P, ACTH, Cor in serum, to investigate the treatment of kidney-yang asthenia syndrome of compatibility mechanism of shenqi pill "reinforcing yang from yin". According to the theory of Chinese medicine that "lung and kideny aioi" and "mother and child phase and", to detect the content of TNF-a in serum of kidney-yang asthenia syndrome COPD rats, to analyze the application that the treatment of kidney-yang asthenia syndrome pulmonary function disease of shenqi pill "reinforcing yang from yin", investigating the treatment of kidney-yang asthenia syndrome of compatibility mechanism of shenqi pill "reinforcing yang from yin" further.MethodsTo dismantle the whole shenqi pill into warming yang group, nourishing yin group, warming yang and nourishing yin group. The warming yang group is composed with aconite root, cinnamon, hoelen, alisma rhizoma and moutan cortex. The nourishing yin group is composed with rehmannia, asiatic cornelian cherry fruit, dioscorea root, hoelen, alisma rhizoma and moutan cortex. The warming yang and nourishing yin group is composed with aconite root, cinnamon, rehmannia, asiatic cornelian cherry fruit, dioscorea root, hoelen, alisma rhizoma and moutan cortex that is the whole shenqi pill group.To divide the SD male rats into normal group, model group and dismantled shenqi pill group randomly. Inject hydrocortisone to the rest rats at the dose of25mg/kg/d for12day, except the normal group. And began to intragastric administration afterwards, supple the normal and model group with liquor natrii chloridi isotonicus in the samme volume, lasting20days.1st day of modeling and1st and21th day of intragastric administration, to weight all the groups of the rats’weight.11th day of modeling and10th and20th day of intragastric administration, to measure all the rats’rectal temperature.10st day of modeling and19th day of intragastric administration, to record in all the rats’swimming time. To anesthetize the rats, get blood from the orbital at21th day, separate the serum and detect the content of PRL, FSH, LH, E2, T, P, ACTH, Cor in serum by the method of ELISA. And the weight of testis, epididymis, prostate, seminal vesicle and adrenal are weighed to detect the index of the weight of testis, epididymis, prostate, seminal vesicle and adrenal. And then get the testis and adrenal tissue fixed in10%formaldehyde, sliced by paraffin, to observe the pathological changes by HE staining.To divide the KM male mices into normal group, model group and dismantled shenqi pill group randomly. Inject hydrocortisone to the rest rats at the dose of0.25mg/kg/d for12day, except the normal group. And began to intragastric administration afterwards, supple the normal and model group with liquor natrii chloridi isotonicus in the samme volume, lasting20days. To record in the mices’spontaneous activity at12st day of modeling and20th day of intragastric administration, the antifatigue and hypoxia tolerance test time at21th day of intragastric administration.To divide the SD male rats into normal group, model group and dismantled shenqi pill group randomly. Inject hydrocortisone to the rest rats at the dose of25mg/kg/d for12day. From1st day to28th day of modeling, to take the rest rats into the glass fume cylinder, fuming40minutes and16cigarettes a day except the normal group.1st and14th day of modeling, to supple the normal group with200μ1liquor natrii chloridi isotonicus by trachea, the rest groups with200tig/200μl lipopolysaccharide binding liquor natrii chloridi isotonicus. Began to intragastric administration at29th day, to supple the normal and model group with liquor natrii chloridi isotonicus in the samme volume, lasting20days. To anesthetize the rats, get blood from the orbital at21th day, separate the serum and detect the content of TNF-αin serum by the method of ELISA.ResultsIn all dismantled shenqi pill group, the results of assay indicated that body weight, organ weight index, rectal temperature, spontaneous activity, antifatigue and hypoxia tolerance test, warming yang and nourishing yin group improve the symptoms better than warming yang group or nourishing yin group. Warming yang group and nourishing yin group could improve the above symptoms also, but the effects were worse than warming yang and nourishing yin group, possessing statistics meaning.The results of assay indicated that the testis and adrenal tissue of observing the pathological changes by HE staining, normal group testis tissue seminiferous tubules structure clearly, the presence of spermatogenic cells at all levels, normal morphology, stromal cells have no obvious abnormalities. Model group testicular seminiferous tubules structure clearly, spermatogenic cells degenerate significantly and the number of them reduced. Compared with model group, warming yang group, nourishing yin group and warming yang and nourishing yin group testicular seminiferous tubular structure clearly, spermatogenic cells degenerate no significantly and the number of them incresed, especially the warming yang and nourishing yin group. Adrenal divids into cortex and medulla, normal group cortex thicker, it can be a clear distinction among the zona glomerulosa, zona fascicular and zona reticularis. The cell structures of zona glomerulosa and zona fascicular are clear, arranged regularly. Model group adrenal cortex is thinner, zona glomerulosa and zona fascicular getting narrower, the structures are unclear. The cells arrangement of them are irregular, skrinkage, staining lighter, cytoplasmic vacuolization and stromal cells getting wider. Warming yang group, nourishing yin group and warming yang and nourishing yin group adrenal cortexs are thicker, the zona fasciculars accounting for the proprotion of the entire cortexs are similar to the normal group. The cells arrange in order, close to the normal group, warming yang and nourishing yin group especially.The results of assay indicated that the content of PRL, FSH, LH, E2, T, P, ACTH, Cor, TNF-a in serum, compared with normal group, the content of T, ACTH and Cor decreased significantly, PRL, FSH, LH, E2, P and TNF-a increased significantly in model group, possessing statistics meaning. Compared with model group, the content of PRL, LH, P and TNF-α decreased significantly in warming yang group, TNF-α decreased significantly in nourishing yin group, T, ACTH and Cor increased significantly and PRL, FSH, LH, E2, P, TNF-a decreased significantly in warming yang and nourishing yin group, all these possessing statistics meaning. Compared with warming yang group, the content of ACTH increased significantly in warming yang and nourishing yin group. Compared with nourishing yin group, the content of PRL decreased significantly and ACTH increased significantly in warming yang and nourishing yin group, possessing statistics meaning.ConclusionThe thought of shenqi pill "reinforcing yang from yin" compatibility is that a large munber of rehmannia, asiatic cornelian cherry fruit, dioscorea root, hoelen, alisma rhizoma and moutan cortex compatibled with a small amount of aconite root, cinnamon. To observe the results of the experiment, the author infers that, to increase the content of T, ACTH, Cor and decrease the content of PRL, FSH, LH, E2, P, TNF-a in sernm are likely to be the pharmacodynamic mechanism that the treatment of kideny-yang asthenia syndrome of shenqi pill "reinforcing yang from yin". In clinical, to decrease the content of TNF-a in sernm is also likely to be the pharmacodynamic of shenqi pill "reinforcing yang from yin" of the treatment of kideny-yang asthenia syndrome pulmonary function disease.
